ObjectiveTo investigate the effects of Tripterygium glycoside (TG) pretreatment on the nerves'regeneration after the transplantations.Methods1. 30 pieces of sciatic nerves (length of 15mm) were offered by male and healthy Wistar rats (15). Forty male SD rats were divided randomly into four groups (n=10, 10mm gap of SD rats'sciatic nerves were repaired by Wistar rats').2. Group A(with TG's pretreatment): 10 pieces of Wistar sciatic nerves(length of 15mm) were dived into TG's pretreatment(400mg/L) for 24 hours in 4 centegrade, and repaired the gaps of SD rats'sciatic nerves about 10mm long. Three control groups were set: Group B: The fresh Wistar sciatic nerves were repaired the gaps of SD sciatic nerves and TG 5mg/(Kg*d) were taken by mouth after transplantations for five weeks. Group C: The fresh sciatic nerves for transplantation were of their own. Group D: The fresh Wistar sciatic nerves were repaired the gaps of SD sciatic nerves.3. Sciatic functional Index(SFI) was measured in the 2,4,8,14 week, gross and microscopic observation were performed termly, electric neurophysiology, transmission electron microscope and regrowed axons were analysed in the 14 week after operation.Results1. SFI recovered in Group A, B and C, and C is more rapid than A and B, but there were no significant differences among them in the 14 week. Compared with Group A, B and C, there were evidently differences among Group D and them (P<0.05).2. The acute immunological rejection of Group A and B were bigger than Group C and easier than Group D.. The inflammatory cells invaded obviously in Group D with axonal degeneration and myelinolysis. There were no significant differences among Group A, B, C in nerve conductive velocity and action potential peak (P<0.05). And bigger than Group D.3. There were no significant differences in the thickness and distribution range of endoneural sheath and lamellar sheath, the area,total number and the thickness of regenerated myelinated nerve fibers(P>0.05). There was almost no regeneration of nerve fiber in Group D.Conclusion1. TG could suppress the acute immunological rejection efficiently after transplantation and promote the regeneration of peripheral nerves.2. TG pretreatment could substituted oral application TG after the nerves transplantations and obviated the TG's adverse reaction.
