| objective:to determine if DNA from Actinobacillus actinomycetemcomitans which are associated with periodontal disease could affect the immune response to other bacterial antigens in the oral cavity, to discuss the feasibility of DNA from Actinobacillus actinomycetemcomitans which are associated with periodontal disease as oral vaccine adjuvants.Method: Cultured Actinobacillus actinomycetemcomitans associated with periodo- ntal disease and extracted DNA.27 rats of common level,were divided into 3 groups randomly:rats were injected with alum(groupâ… ),alum-GTF(groupâ…¡),or alum-GTF together with DNA(groupâ…¢),then serum IgG antibody were tested in the 0,1,3,6weeks in ELISA. One-way analysis of vaccine and the Student- Newman- Keuls(SNK) multiple comparison test were used to analyze differences among groups.Statistical singificance was assumed with probability values of less than 0.05.RESULTS: In the first week,third week and sixth week, The groups receiving alum-GTF demonstrated significantly increased IgG antibody levels compared to the alum,the groups receiving alum-GTF plus DNA demonstrated significantly reduced IgG antibody levels compared to the alum-GTF alone group.Conclusion: Actinobacillus actinomycetemcomitans DNA associated with periodo- ntal disease as adjuvant with host response to GTF has the potential to reduce the effectiveness of natural or administered dental caries and periodontal disease vaccine. |
