Objective:To investigate the expression and distribution of inhibinαprotein in th serous carcinomas of ovary. To research whether the expression of inhibinαmRNA and protein would be increased when the human ovarian cancer cell-SKOV3 were chemotherapy by Cisplatin(DDP). We expect that the study will help us to elucidate the role of inhibinαin the formation of the ovary cancer.Methods:1,The expression of inhibinαwas examined in 30 normal ovary tissue, 30 nonmalignant cystadenomas ovarian serous tumors and 45 malignant ovarian serous tumors by EliVision immunohistochemistry . 2,We further to detect the change of inhibinαwith cell chemotherapy technique on the base of research of clinical specimen ovary serous cancers. 2.1,We cultivated cell SKOV3 of human ovarian serous cancers . 2.2,We detected the cell growth inhibiting of SKOV3 with different concentration of DDP and at the different time by MTT colorimetry, so we could select the best DDP concentration. 2.3,We examined the expression change of inhibinαmRNA after the DDP affecting the SKOV3 by RT-PCR. 2.4,We examined the protein expression change of inhibinαafter the DDP affecting the SKOV3 by immunochemistry staining.Results:1,Immunohistochemistry experiments showed that inhibinαprotein expressed on the normal ovary tissue, nonmalignant ovarian serous tumors and malignant ovarian serous tumors ,the total positive of inhibinαimmunostaining were 93.3%,46.7% and 24.4%,respectively.,there were significant difference between three groups(P<0.05).The positive expression of inhibinαprotein on malignant ovarian serous tumors was significantly lower than the nonmalignant ovarian serous tumors and the normal ovary tissue(P<0.05). 2.1,We found that the effect of cell growth inhibiting of SKOV3 was best , when the DDP concentration was 12.5umol/L after dealing the cell SKOV3 48 hours ,with different concentration of DDP(200,100,50,25,12.5,6.25,3.125,1,5625umol/L) and at the different time(12h,24h,36h,48h) by MTT colorimetry. When the DDP concentration exceeded 12.5umol/L, along with increasing of the DDP concentration, the cell apoptosis was increasing. when the DDP concentration was low 12.5umol/L , the cell inhibition was not obvious. The optimal drug density is 12umol/L according to IC50. 2.2,The results of RT-PCR showed that the expression of inhibinαmRNA also gradually increased as the time increasing ,When the DDP concentration was 12umol/L to deal SKOV3. And when the DDP dealt the cell SKOV3 48 hours, the expression of inhibinαmRNA was significantly high than than control group(P<0.05), other time groups(4h,8h,12h)contrasted the control group, but the difference has no significance( P>0.05). 2.3,Immunohistochemistry experiments showed that when the DDP concentration was 12umol/L to deal SKOV3, as the time increasing , inhibinαprotein started to appear after DDP affecting SKOV3 24 hours, the expression of inhibinαprotein was significantly high than than control group(P<0.05), but the 48h group was no difference to the 24h group(P>0.05);Other time groups (4h,8h,12h)contrasted to the control group, the difference has no significance( P>0.05).Conclusions:1,The results of clinical specimen shows that the expression of inhibinαprotein on the malignant ovarian serous tumors is significantly lower than the nonmalignant ovarian serous tumors and the normal ovary tissue(P<0.05), This shows that the expression of inhibinαprotein may be suppressed in the ovary serous cancers. 2,The results of human ovary cancer cells SKOV3 show that when the DDP dealt the SKOV3, as the time wad increasing , the expression of inhibinαalso increasing by RT-PCR and immunohistochemistry . This suggests that the maglinant ovarian serous tumor cells enhance the secretion of inhibinαafter chemotherapy. 3,These two study including clinic and experiment indicate that inhibinαmay be possibly a tumor inhibiting fator in the ovary serous cancers.
