| Objective: To investigate the protective effects on hepatic ischemia reperfusion injury (IRI) and the impact on the apoptosis and the expression of TNF-α, IL-1, HIF-1α, Caspase-3 by panax notoginseng saponins(PNS) preconditioning in rats after partly hepatectomy and liver autoplastic transplantation.Method: Establish partly hepatectomy and liver autoplastic transplantation model of rats .54 cases of male Sprague-Dawley rats were randomly divided into three groups according to the different drugs administrated before liver reperfusion: 18 rats in panax notoginseng saponins preconditioning group((PNS), 18 rats in control group with aqua pro injection pretreatment(CM) and 18 rats in sham-operated group(S). The animals were killed at 2h, 6h, 24h after autoplastic transplantation liver reperfusion or sham operation respectively. Plasma samples were collected for ALT and AST test; Part of the liver tissue were made in paraffin-embeded speciments to detect rats liver histological change(HE), apoptosis (TUNEL) and the protein expression of TNF-α, IL-1, HIF-1αand Caspase-3 by immunohistochemical.Results: (1)At 2h, 6h, 24h after liver autoplastic transplantation reperfusion , the levels of ALT,AST and apoptosis index were higher in the group of CM and PNS than the group of SO significantly(P<0.01);(2)The levels of ALT,AST and apoptosis index and the expression of TNF-αprotein ,IL-1 protein and Caspase-3 protein were higher in the group of CM than the group of PNS significantly(P<0.01); (3)The expression of HIF-1αprotein were higher in the group of PNS than the group of CM significantly at 2h, 6h, 24h after auto-transplantation liver reperfusion (P<0.01).Conclusion: (1) Partly hepatectomy liver autoplastic transplantation can lead to the severe hepatic ischemia reperfusion, which may be associated with the augment of oxyradical, cytokine levels and apoptosis. (2) Panax notoginseng saponins(PNS) preconditioning can attenuate liver ischemia reperfusion injury (IRI)in rats partly hepatectomy liver autoplastic transplantation. (3)The antiapoptotic effects of PNS may be associated with inhibiting the expression of genes of TNF-αand Caspase-3. (4) The expression up-regulation of HIF-1αcan decrease the levels of TNF-αand IL-1, which can lessen ischemia reperfusion. |
