Objective:To establish the blood glucose fluctuation rat model,and detected the level of ceramide of the serum and blood vessels in diabetic rats and blood glucose fluctuation diabetic rats model.Method:The Male Sprague-Dawley(SD) rats were randomly assigened into two groups:normal group rats(n=20) and model group rats(n=40).Normal group rats were fed with normal diet,model group rats were fed with a high-sucrose-high-fat diet and after 6 weeks a low dose of Streptozotocin(STZ,35mg/Kg) was injected to induce hyperglycemia.And then the model group rats were divided randomly into two groups again:diabetic group rats(n = 20) and blood glucose fluctuation group rats(n = 20).Blood glucose fluctuation group rats were induced by Subcutaneous injecting insulin twice daily.All rats were executed after intervene 3 months,metabolic cage collect 24-hour urine one week before all rats were executed.The HbA1c,TG,TC,LDL,HDL,ALT,AST,BUN,Cr were measured.Extracted lipid from serum and arteria aorta to detect the level of ceramide by HPLC-MS method.Results:1.The SD rats were successed induced into the diabetic model by fed with a high-sucrose-high-fat diet and a low dose of STZ(35mg/Kg) was injected,and all diabetic model rats blood glucose exceeded 16.7mmol/l.2.By the method of twice-daily subcutaneous insulin injection,the average blood glucose levels(MBG),the standard deviation of daily average blood glucose(SDBG) and the largest amplitude of glycemic excursions(LAGE) were significant differences(P<0.05) in the compare of normal group,diabetic group and blood glucose fluctuation group.3.HbA1c,TG,TC,LDL,HDL had no differences between diabetic group rats and blood glucose fluctuation group rats(P>0.05),but raised compare normal group rats(P<0.05).Three groups rats had significant differences(P<0.05) in the ALT,AST,BUN,Cr.4.Normal group,diabetic group and blood glucose fluctuation group had significant differences in the ceramide level of serum and arteria aorta(P<0.05).Conclusions:1.The blood glucose fluctuation diabetic rats model are successed induced by he method of twice-daily subcutaneous insulin injection in Diabetic rats.2.Blood glucose fluctuation can aggravate Diabetic rats Liver function and renal function.3.The level of ceramide of the serum and blood vessels in diabetic rats is increased and blood glucose fluctuation makes it more evident.
