ObjectiveThe objective of the study is to investigate the roles and effects of garlicin on cultured human periodontal ligament cells in vitro,in order to clarify the influence of garlicin on the host cell cycle mechanism for clinical use.Methods1.hPDLCs derived from 12-20 years old volunteers due to the need for removal of post-orthodontic periodontal ligament cells(hPDLCs), identified by the train after the 5th passage cells for research.2.The division of experimental control group,the standard calcium hydroxide in the control group(saturated solution of 1%) and allicin (3,6,9 ug/ml) concentrations of the three test groups.3.The use of flow cytometry(FCM) in vitro is to observe and analyze the phase changes of DNA synthesis and cell cycle in human periodontal ligament cells(hPDLCs) after different garlicin concentrationsResults:1.The tissue culture is a efficient method for primary hPDLCs culture identified by immunohistochemistry.The S-shaped growth curve of HPDLCs are in line with the general rules.2.By calcium hydroxide(saturated solution of 1%) and different concentrations of garlicin treatment,hPDLCs' cell cycle in G1 phase phase(DNA synthesis of pre-),in addition to calcium hydroxide group and Garlicin 6ug/ml group,was statistically significant among the differences in each groups.(P<0.05);in G2 phase(DNA synthesis of the latter),the differences between the calcium hydroxide group and other groups were statistically significant(P<0.05),Garlicin 3ug/ml group and control group,as well as the differences between Garlicin 6ug/ml group and 9ug/ml group were not statistically significant;in S phase (DNA synthesis phase),the differences between each group were statistically significance(P<0.05);PrI(Proliferation Index)value,which reflects the proliferation activity of proliferation index,in addition to calcium hydroxide group and the Garlicin 6ug/ml group,was statistically significant among the differences in other groups.Conclusion1.Different concentrations of allicin and calcium hydroxide on human periodontal ligament Cells the role of cell cycle block.2.Different concentrations of allicin and calcium hydroxide can human periodontal ligament cells in S phase block.3.Different concentrations of allicin on human periodontal ligament cells block was stronger than calcium hydroxide.
