Objectives(1) To observe the differential expression of Skp2 and P27kip1 between normal skin and the lesions of psoriasis vulgaris;(2) To investigate the relationships between Skp2 and P27kip1 in the lesions of psoriasis vulgaris,and study the effects of them in the pathogenesis of psoriasis.Methods(1) The normal skin and the lesions of psoriasis vulgaris was divided into two parts separately, one was put into 10% formalin solution to prepare for immunohistochemistry ,another was put into liquid nitrogen to prepare for reverse-transcription polymerase chain reaction (RT-PCR);(2) The expression of Skp2 mRNA and P27kip1 mRNA were investigated in 10 cases of normal skin and 30 cases of psoriasis vulgaris using reverse-transcription polymerase chain reaction (RT-PCR);(3) The expression of Skp2 protein and P27kip1 protein were investigated in 10 cases of normal skin and 30 cases of psoriasis vulgaris using immunohistochemistry method.Results(1)The expression of P27kip1 protein significantly decreased in the lesions of psoriasis vulgaris compared with normal control group (P<0.01). but there was no difference of the expression of P27kip1mRNA between psoriasis vulgaris and normal control group (P>0.05);(2)The expression of Skp2 mRNA and protein were significantly increased in the lesions of psoriasis vulgaris compared with normal control group (P<0.05);(3) P27kip1 protein expression was negatively correlated with Skp2 protein in the lesions of psoriasis vulgaris (P<0.01).Conclusions1,The overexpression of Skp2 mRNA and protein and the decreased expression of P27kip1 protein may play an important role in pathogenesis and development of psoriasis vulgaris;2,Skp2 may reduce the protein level of P27kip1 to promote the proliferation of keratinocytes in psoriasis vulgaris.
