| [Objective]1.To research the effect ofβ-elemene at different concentration on apoptosis of human gastric cancer BAC823 cells in vitro.2.To research the effect ofβ-elemene on P38MAPK at the time of apoptosis of human gastric cancer BAC823 cells.To infer the regulating action of signal transduction pathway P38MAPK during apoptosis of cancer cells induced byβ-elemene.[Methods]Human gastric cancer BAC823 cells were cultured in vitro.The whole experimental process was divided into three batches.In each batch,the cells were divided into two groups including blank control group and drug groups.In terms of concentration,drug groups were divided into 0.01mg/ml, 0.02mg/ml,0.04mg/ml,0.08mg/ml and 0.10mg/ml,0.16mg/ml group.The effect time was 24h.1.Inhibition of proliferation was measured with MTT assay.2.Apoptotic rate and fraction of the cell cycle phase were measured with flow cytometry(FCM).At the same time,morphological and adhesive changes of the cells were observed under an inverted microscope.3.The level of P-P38MAPK was measured with immunocytochemical method. [Results]1.The result of MTT examination showed there was positive correlation between drug concentration and inhibition of proliferation.(P<0.01) And it was estimated IC50 of BAC823 cells was 0.078mg/ml withβ-elemene after 24h.2.The result of FCM examination and morphological observation showedβ-elemene can inhibit BAC823 cells at S phase and induce apoptosis.But between the rate of apoptosis and the drug concentration,there is not line correlation.3.The result of SABC showedβ-elemene enhanced the level of p-p38MAPK and it was enhanced with the drug concentration increased. (P<0.01)[Conclusions]1.β-elemene can inhibit the proliferation of cancer cells and there is posstive correlation between the effect and the drug concentration.2.β-elemene can induce apoptosis of cancer cells.At certain range apoptotic rate raised with the drug concentration increased,and beyond the range apoptotic rate will decend,necrosis increasing.3.β-elemene can active P38MAPK pathway and the pathway probably take part in regulating the apoptotic process. |
