OBJECTIVE:To investigate the relationship between the 6-Keto-PG F1α,TXB2 and the endotheliosis and thrombosis of the new endomembrance on the prostheses seeded by CD34+ stem cellsMETHOD:Twelve dogs were randomized into two groups.Prostheses covered with ePTFE or Dacron were implanted into the abdominal aorta(AA) and inferior vena cava(IVC).in each group,6 dogs were implanted by prostheses seeded by CD34+ stem cells,4 dogs were implanted by autogenous blood only as control.The blood was collected in Preoperative and 3,7,14,30,60 days postoperative from femoral vein.Light and electron microscopy were applied to observe the endotheliosis and thrombosis of the new endomembrance when harvesting all the prostheses in the 60 days.RESULT(1)The platelet concentration in the experimental group increased at first,then decreased and maintained at a certain level,while the control group, after a marked increase,remained at a high level;(2)The serum PGF1αin experimental group reduced firstly and then increased while the control group decreased significantly and then maintained at a lower level;(3) The TBX2 concentration in the experimental group increased at first, then decreased and maintained at a certain level,while the control group,after a marked increase,remained at a high level;(4) The serum P/T ratio in experimental group reduced firstly and then increased while the control group decreased significantly and then maintained at a lower level;(5)The neointimal of prostheses in the control group is hyperplastic significantly and has thrombosis.CONCLUSION:(1) It can inhibit the adhesion,aggregation and activation of platelet,also inhibit the production of bioactivators,such as growth factor, cytokines,adhesion molecules and chemokines through displacing blood vessel with prostheses seeded with CD34+ stem cell. (2)It makes endothelial cell grow faster,which can produce prostacyclin, adenosine,nitric oxide,VEGF and other bioactivators through displacing blood vessel with prostheses seeded with CD34+ stem cell.And then they inhibit PGF1αand P / T ratio of over-reduced effectively,restrain TXB2 and platelet excessive rise,finnally prevent intimal hyperplasia and thrombosis.
