Objectives: To observe the gene expression of VEGF and proliferation of Ishikawa cells which is treated by combining RNA interference with oxymatrine in endometrial carcinoma.Methods: The plsamids of RNA interference were designed and constructed on the basis of pRNAT-CMV3.2-Neo Vector. pRNAT-CMV3.2-Neo-VEGF was constructed to silence VEGF gene, while pRNAT-CMV3.2-Neo-neg was the negative control. Then the plasmids were transfected into Ishikawa cells,and oxymatrine was injected to the cells. The results were observed by semiquantitative RT-PCR, Western blotting, MTT and FCM, and morphological changes of cells were also observed . The effect of invasion was evaluated by using Transwell chambers attached to Matrigel.Results: The plsamids of RNA interference were transfected into Ishikawa cells successfully. RNA interference combined with oxymatrine could effectively inhibit the proliferation of Ishikawa cells, induce apoptosis and down-regulate the expression of VEGF in the level of mRNA and protein. FCM analysis proved that cell population in G1 phase was increased and cell population in S phase was decreased . The morphology was changed and the invasion of tumor cells to basement membrane was significantly suppressed.Conclusions: RNA interference combined with oxymatrine effectively inhibited the proliferation and invasion of Ishikawa cells ,and induced apoptosis and differentiation, which might be related to the down-regulation of the expression of VEGF in the level of mRNA and protein.It is the first time to combine RNA interference with oxymatrine to inhibit VEGF in the therapy of endometrial carcinoma. It proved that the effect of RNA interference combined with oxymatrine is more effective than single gene therapy and single drug therapy, which provides a new approach to other tumors.
