The Expressions Of CD28 And CTLA4 On T Lymphocytes Of Bone Marrow And The Changes Of TNF-α And IFN-γ Treated By Xuefusheng Decoction In Aplastic Anemia Mice Model

Objective:To explore the possible mechanism of chinse medicine-xuefusheng decoction in the treatment of aplastic anemia,Immune-induced aplastic anemia model mice was established,the expression of CD28 and CTLA4 on T Lymphocytes of bone marrow of aplastic anemia mice was detected,the levels of tumor necrosis factor-α(TNF-α) and interferon(IFN-γ) of plasma and supematant obtained from spleen Lymphocytes by culture containing PHA were measured.The changes by treatment of xufusheng decoction were investigated.Method:Immune-induced aplastic anemia model mice was established,fifty mouse were randomly divided into five groups,normal control,model control,the treatment of xuefusheng decoction,the treatment of xuefusheng decoction and CyclosporinA(CsA),the treatment of CyclosporinA(CsA),ten mouse every group. The counts of peripheral blood cells and marrow mononuclear cells were detected,the bone marrow's pathology were observated,marrow mononuclear cells were exposured by culture containing PHA,the expression of CD28 and CTLA4 on T Lymphocytes at 0 h and 48 h after PHA exposure were analyzed by two color flow cytometry.The levels of tumor necrosis factor-α(TNF-α) and interferon(IFN-γ) of plasma and supematant obtained from Spleen Lymphocytes by culture containing PHA were measured by double antibodyes sandiwich -ELISA.Results:Comparing the group of xuefusheng decoction to model group, xuefusheng decoction and CyclosporinA(CsA) to CyclosporinA(CsA),the counts of peripheral blood cells and marrow mononuclear cells were significantly elevated (P＜0.05).the degree of bone marrow proliferation has improved evidently,hematopoietic cells have increased significantly(P＜0.05),The expression of CD28 in the group of xuefusheng decoction were obviously lower than that of model group,but the significant difference between the groups of xuefusheng decoction and CyclosporinA(CsA) and CyclosporinA(CsA) were not found,Comparing the group of xuefusheng decoction to model group,xuefusheng decoction and CyclosporinA(CsA) to CyclosporinA(CsA),the expression of CTLA4 were no statistical significance.The levels of tumor necrosis factor-α(TNF-α) and interferon(IFN-γ) in the supematant obtained from Spleen by culture containing PHA were obviously lowered.Concision:The treatment of xuefusheng decoction can be effective to regulate the immunological disorder in aplastic anemia,recover the hematopoietic function and enhance the effect of the CyclosporinA(CsA) therapy in AA.
