The Sphincter of Oddi Dysfunction (SOD) is the syndrome that has a group of mainly clinical manifestations, such as recurrent upper abdominal pain, jaundice, increased amylase of blood or urine. SOD often occurred in patients after cholecystectomy, it is also called post-cholecystectomy syndrome. Current study confirmed that there was neural reflex and humeral regulation between gallbladder and SO. Gallbladder tension and normal systolic and diastolic movement have significantly impact on the movement of SO. Moreover, the inhibition of SO'movement by the CCK peak after prandial disappeared, it shows whether gallbladder existence or not plays an important role in the regulation of SO by the level of body fluids. SO the abnormal regulation of nerves and body fluids after cholecystectomy maybe one of the factors of that SOD always occurred in the patients after cholecystectomy. Now, all the studies of homeland and abroad are focusing on the regulation of SO and pathways regulating in the SO cells, while there was little research on the change of SO itself. SO we need to establish animal model of post-cholecystectomy to fatherly study. With regards of these, we should detect the contraction movement and EMG of the rabbits'SO in vitro after cholecystectomy, determinate the myosin heavy chain expression in the SO, which reflects the ability of contraction, and watch the smooth muscle cell's change by transmission electron microscopy in SO of rabbits after cholecystectomy. By which, we can explore whether adaptive changes have taken place in SO after cholecystectomy, by which we can fatherly study how to block the occurrence and development of SOD.ObjectiveTo study the alteration of rabbits' sphincter of Oddi after cholecystectomy. Methods68 New Zealand rabbits were divided into four groups randomly: group A, B, C and D. In group A, rabbits were only subjected to laparotomy and brood for 4 weeks, while in group B,C and D, rabbits were subjected to cholecystectomy and brood for 2, 4 and 8 weeks respectively. When time is up, Then the rabbits'sphincter of Oddi were dislodged, 6 rabbits of A and D groups were detected by myosin heavy chain(MHC) immunohistochemistry and 12 rabbits of all groups were gotten manometer and EMG testing in vitro and MHC expression examination by western blot. And 4 rabbits in both group A and group D were examined by transmission electro microscope.Results1. Amplitude of manometer wave increased significantly overexpressed in B,C,D groups, but A group (P<0.05), MHC expression in SO of B, C, D groups had gradually increased compared with the A group and had significant difference(P<0.05) .2. Immunohistochemistry showed smooth muscle cells of D group were more concentrated than A group in the dye of myosin. the protein expression of MHC increased at 2, 4, 8 week after cholecystectomy (P <0.05).3. Dense bodies in smooth muscle cells increased in group D compared with group A.Conclusion1. The motor function of SO was enhanced after cholecystectomy.2. The transmission electron microscopy images of SO cells changed after cholecystectomy3. The protein expression of MHC increased after cholecystectomy...
