ObjectiveMyocardial fibros(MF) is one of the major appearances of left ventricle reconstitution in hypertention.,in morphous,left ventricular hypertrophy is the macroappearance,multcollogen deposit,each type of collogen disproportion and disorder is the microappearance,the development of MF can produce many serious complications,for example,cardiac failure,cardiac dysrhythmia and cardiogenic sudden death,and so on,in the cell factors of effect MF,it is validated that transforming growth factor(TGF-β1) has the most closed relation with ECM,it is the received healing target of organ fibrosis,in recent report,connective tissue growth factor(CTGF) is one of the CCN family members,a good deal of studies of CTGF are used in the fibrosis of liver, kigney,lung and so on,it has many homoplastic biologrical function with TGF-β1,up to now,CTGF is presumed the downstream effect mediator of TGF-β1,obantibody of CTGF can inhibit the fibrotic effect that been induced by TGF-β1,but,at present,it has no exact report intermal that whether or not CTGF participate the myocardial of hypertension process,The object of the expreriment is to investigate roles of the connective tissue growth factor(CTGF) in the development of myocardial fibrosis and left ventricular hypertrophy in renovascular hypertensive rats.Methods(1)Subgroup:Use healty masculine SD rats,weight 180-220g,age 12 weeks;The experimental rats were divided into two groups randomly,experiment group and control group,the experiment group have 20 rats and the control group have 10 rats;the experiment group were established into renovascular hypertensive rat models,and the control group are carried out the same operation progress,but not been settled down silver clip,so the renal artery is not stenosal.Then the two group rats are breeded eight weeks.To the forth week,there are eight rats's blood pressure did not reach to the goal in the experiment group,so they can not enter the ulterior experiment. Because postoperation infection,to the eighth week,one rat die in the experiment group and one rat die in the control group.(2)Determine LVWI and EF%of the two group rats by 2-Doppler echocardiography.(3) Take the cardiac musclular tissues of the connecting piece of transverse section in the left artrium,use immunohistochemical methed to semiquantitative analysis the distribution and expression of CTGF,TGF-β1 in the two group's left atrium muscle.Use SP staining methed to observe the morphous of left artium muscle collogen,image analysis and measure CVF systematically.(4)Statistical treatment:use SPSS11.5 monfactor variance analysis,q check and correlation analysis.All measurement data are expressed by means of mean±standard deviation.P<0.05 denote that the difference has significance,P<0.01 denote that the difference has polisignificance.Results(1)The SBP is higher in experiment group than in control group obviously.(2) Measure LVWI and EF%of rats by 2-Doppler echocardiography,experiment group have significant difference with control group.(3)The expression of CTGF and TGF-β1 in the myocardium of left ventricle in experiment group is booster obviously than in control group(P<0.05).(4)Correlation analysis indicate that CTGF and TGF-β1 assume positive correlation.(r=0.626,P<0.05).Conclusion(1)SBp of renal vascular hypertension rats step up obviously.(2)The measure value of 2-Doppler echocardiography show left ventricular wall hypertrophy,left ventricular chamber shrink,LVMI increase highly.(3) CTGF and TGF-β1 mainly express in interstitial substance of cardiac mulscle.CTGF and TGF-β1 in interstitial substance of cardiac mulscle of renal vascular hypertension rats express higher in experiment group than in control group obviously.It is thus clear that there is a great deal of buffily particulate matter is expressed as CTGF in experiment group rats,the expression of TGF-β1 submits powerful positive,while,in control group it is thus clear that there is a few helvolus particulate matter is expressed as CTGF in control group rats,the expression of TGF-β1 submits weakly positive.The connective tissue growth factor and TGF-β1 may play important roles in the development of myocardial inter stitial fibrosis.
