Objective:To study the effect and mechanism of Janus Kinase2-selective inhibitor AG490 in proliferation,apoptosis and the capacity of invasion and metastasis of human cervical cancer Hela cells.Methods:(1) We cultured the human cervical cancer cell line Hela cell,Observated the growth of cells and drawed the growth curve mapping of cell.(2) Essay grouping:Hela cell were cultured and treated by AG490 with 25μmol / L,50μmol / L,100μmol / L concentrations respectively.(3) Hela cell were cultured and treated by AG490 with 25μmol / L,50μmol / L,100μmol/L concentration for 24,48,72 hours,the effect of AG490 on Hela cell were analyzed by MTT assay,respectively.(4)Hela cell were cultured and treated by AG490 with 50μmol/L concentration for 24,48,72 hours,the apoptotic rate and cell cycle distribution were analyzed by Flow cytometry.(5) Western blot were performed to detect the expression of STAT3,P - STAT3, MMP-2,VEGF proteins.Results:JAK inhibitor AG490 inhibited the cell proliferation in a dose, time-dependment manner;AG490 also induced apoptosis in Hela cell.The apoptosis cell increased with the elapse of time;AG490 inhibited G1 to S cell cycle transition and induced G1 cell cycle arrest in Hela cell;AG490 result in significant downregulation of P- STAT3,MMP-2 and VEGF expression in Hela cell line.Conclusion: 1.The activation of JAK/STAT signaling pathway continued in cervical cancer Hela cell lines.2.AG490 inhibited the proliferation of Hela cell in a dose,time-dependent manner.3.AG490 inhibited G1 to S cell cycle transition in a time-dependment manner and induced G1 cell cycle arrest in Hela cell,AG490 induced the apoptosis cell increased with the elapse of time.4.AG490 can down-regulate protein expression of P-STAT3,MMP-2 and VEGF in cervical cancer Hela cell,inhibit the capacity of invasion and metastasis of cervical cancer Hela cell.5.Blocking JAKs/STATs signaling pathway may become a novel strategy in treatment and prevention of invasion and metastasis of cervical cancer.
