To discover the chemical consitituents in TCMs is of great importance for further identifying active components of TCMs responsible for the pharmacological effects and for constructing scientific quality control standards of Chinese patent drugs.High Performance Liquid Chromatography /Mass Spectrometry(HPLC/MS) has a very powerful capability for separation and structural identification,which can characterize chemical composition and content changes of the complex system quickly and easily. Hence,it has become an indispensable analytical tool in the study on chemical constituents of TCMs.In this thesis,HPLC-DAD-ESI-MS~n and HPLC/Q-TOF-MS based method were proposed in the studies on the chemical analysis of "NAO-DE-SHENG" tablet and "WEI-FU-CHUN" tablet,and HPLC-DAD-ESI-MS~n was also employed in the analysis of rat plasma samples after oral administration of"NAO-DE-SHENG" tablet.The main contents and results of this thesis are listed as follows:1.A HPLC-DAD-ESI-MS~n method was constructed to reveal of chemical substance in "NAO-DE-SHENG" tablet.A total of 43 peaks were detected,and all the herbal peaks were assigned.By comparing retention time,UV absorption wavelength and MS information with standard compounds information and literature data,a total of 23 compounds were successfully identified.2.A HPLC-DAD-ESI-MS~n method was developed to analysis the prototype compounds and their metabolites of rat plasma samples after oral administration of "NAO-DE-SHENG"tablet.In this study,18 prototype compounds and 5 metabolites were detected,and we identified or tentatively elucidated their structures.This method is validated as a simple,reliable method for rapid identification the prototype compounds and their metabolites of rat plasma samples after oral administration of "NAO-DE-SHENG" tablet.3.HPLC-DAD-ESI-MS~n and HPLC/Q-TOF-MS based method were employed to reveal of chemical substance in "WEI-FU-CHUN" tablet.A total of 60 peaks were detected,and all the herbal peaks were assigned.In the end,a total of 46 compounds were identified by comparing retention time,UV absorption wavelength and MS information with standard compounds information and literature data,and also by high-resolution mass spectrometry data analysis and database information retrieval.The method initially defined the chemical substance of"WEI-FU-CHUN" tablet.The research of the present work indicated that High Performance Liquid Chromatography/Mass Spectrometry(HPLC/MS) is indeed a very powerful analytical tool for the studies on chemical substance of TCMs.The results of these studies would be helpful to discover the biologically active compounds and provided valuable information for the development and improvement of the quality control of "NAO-DE-SHENG" tablet and "WEI-FU-CHUN" tablet.
