| Quinone Reductase(QR) is one of the major phaseⅡenzymes that related to chemoprevention.In this research,we aim to find some compounds that can induce QR with natural origins.Firstly,we tests,in vitro,the QR induction of many natural products including Magnolia cylindrica ect.Then,we select Magnolia cylindrica and Salvia miltiorrhiza to make further research.Eight compounds are purified from M.cylindrica,most of them are myrislignans, and thay are:syringin,lariciresinol 4-O-glucoside,conicaoside,(+)-syringaresinol O-β-D-glucopyranoside,lirioresinol A,1-(α-L-rhamnosyl(1-6)-β-D-glucopyranosyloxy)-3,4,5-trimethoxybenzene eleutheroside E and quinquenin L1 accordlingly.By the technology of LC-MS we find three major compounds of the water-soluble compounds in S.miltiorrhiza,they are salvianolic acid B(SaB),lithospermic acid (LA),rosmarinic acid(RA) accordingly,and all of them can react with GSH.However, the QR induction tested in vitro shows all of them have week QR induction activity.The contradiction may been arised from the water-soluble compounds cannot been absorbed by cells.So the structural modification is taken into consideration.After structural modification,we glad to see,the QR induction of LA-nCH3 and RA-nCH3 greatly improved comparing to LA and RA.On the other hand,however,SaB-nCH3 has the same QR induction with SaB.Finally,in cell,LA-nCH3,RA-nCH3 and Sab-nCH3 can been tested by LC-MS,also we find both of LA-nCH3 and RA-nCH3 can react with GSH while Sab-nCH3 cannot. |
