| Objective:To investigate the effect of Ad-ING4,Ad.IL-24 on proliferation and migration of glioma cells and explore its probable mechanism.Methods:U251 were infected with Ad-ING4,Ad.IL-24; Microscope was used for observing the infection efficiency, RT-PCR for examining the transcription level of ING4 and IL-24, MTT and FACS for investigating the effect of ING4 and IL-24 on the proliferation of U251; Boyden chamber motility assay and scrape motility assay for checking the effect of ING4 and IL-24 on the migration of U251; Western Blot for detecting expression of NGF and TrkA; Pull-down assay for detecting expression of active RhoA, and analyzing association between them.Result:All of the methods revealed that ING4,IL-24 express well in U251 cells, inhibit the proliferation and migration of U251 significantly. Moreover, overexpression of ING4,IL-24 result in decreasion of NGF, TrkA and active RhoA.Conclusion:ING4,IL-24 mediated inhibition of the proliferation and migration of human glioma cells was resulted from decreasing expression of NGF, TrkA and active RhoA. |
