Objective:To observe the characteristics of methamphetamine hydrochloride and to study the function of COX-2 protein and PGE2 in the methamphetamine intoxication rats.Methods:Forty male Sprague-Dawley rats were randomly divided into control group(n=10) and experimental group (dividing it into the first, fourth and seventh group after the last injection, n=10) Rats in the experimental group were intraperitoneal injected with methamphetamine hydrochloride (20mg/kg), and those in the control group were injected with saline with the same volume (twice a day at 8:00 and 20:00 respectively for 4 continuous days), then, observing their behavioral changes. Examining the protein expression of COX-2 of intoxication rats in related encephalic region by immunohistochemistry technique and analyzing the test results through image.Results:Both COX-2 and PGE2 are featured with the similar expression changes in the different areas of brain with obviously masculine signals. The expressions of COX-2,PGE2 varied little in the control groups. However, the expressions of COX-2,PGE2 in groups(1d group,4d group,7d group) varied obviously。At the first day, the expressions deduced gradually. At the fourth day, the expressions deduced to the lowest.At the seventh day, the expressions recovered to the normal level approximately. The expressions of COX-2,PGE2 varied little in the control groups.Conclusion:1. Setting up the model of methamphetamine intoxication.2. The abnormal expression of COX-2,PGE2 of methamphetamine intoxication rats in PFC,CPu,CA showed that it took part in the methamphetamine induced neuro- toxicity.3. The abnormal expression of COX-2,PGE2 of rats Sub-acute intoxication of methamphetamine in PFC,CPu,CA showed its close connection with protection of methamphetamine induced neurotoxicity.4. There is a close relationship between COX-2/PGE2 system and MA toxicity.
