Objective:1. Immunohistochemical technique was used to examine the expression of VEGF in colorectal carcinomas, adenomas and normal mucosa samples.Examine the expression of VEGF in colorectal carcinomas and their surrounding tissues and surgical margins.Study its relationship with clinicopathologic of colorectal carcinomas.2. Immunohistochemical technique was used to examine the expression of COX-2,CD34 and count the signal microvessl density of CD34 labeled. Study relationship between COX-2,CD34 and VEGF in colorectal carcinomas.Methods:1. Immunohistochemical technique SP was used to examine the expression of VEGF in 98 cases of colorectal carcinomas,22 cases of adenomas and 15 cases of normal mucosa samples.Examine the expression of VEGF in colorectal carcinomas and their surrounding tissues and surgical margins.2. Immunohistochemical technique SP was used to examine the expression of COX-2 and CD34 in 98 cases of colorectal carcinomas,22 cases of adenomas and 15 cases of normal mucosa samples.Count the microvessl density(MVD) under the microscope.Results:1. The positive expression rates of VEGF in colorectal carcinomas,adenomas and normal mucosa samples were 82.65%,68.18%,26.66% respectively.There existed significant difference of the positive expression rate of VEGF between the groups each other(P<0.05 for both).The positive expression rates of VEGF in colorectal carcinomas,their surrounding tissues and surgical mrgins were 82.65%,65.3%,54.08% respectively.There existed significant difference of the positive expression rate of VEGF between the groups each other (P<0.05 for both).2. The expression of VEGF protein shows no correlation with patient,sex,tumor location(recum or colon), there was no significant difference (P>0.05).VEGF protein expression in well-differentiated adenocarcinoma was 64%,in differentiated adenocarcinoma was 84.6%,in poorly differentiated adenocarcinoma was 94.1%,there was significant difference between the groups each other(P<0.05 for both).VEGF protein expression in subserosa group was 75%, in extraserosal infiltration group was 85.71%,there was significant difference between the groups(P<0.05). VEGF protein expression in lymphnode metastasis positive group was 89.74%, in lymphnode metastasis negative group was 77.96%, there was significant difference between the groups (P<0.01). VEGF protein expression in Dukers A/B group was 78.84%, in Dukers C/D group was 86.95%, there was significant difference between the groups (P<0.05).3. Staining of CD34 was easy to identify for its high specificity,sensitivity and clear back ground.The MVD of colorectal carcinoma was 37.04±10.09,Compared with colorectal adenomas,there was significant difference(P<0.01);Compared with normal mucosa samples,there was significant difference(P<0.01). The MVD of colorectal adenomas was 26.57±14.45, Compared with normal mucosa samples,there was significant difference(P<0.01).4. The positive expression rates of COX-2 in colorectal carcinomas,adenomas and normal mucosa samples were 73.46%,59.09%,20% respectively.There existed significant difference of the positive expression rate of VEGF between the groups each other (P<0.05 for both). The expression of VEGF protein shows no correlation with patient,sex,tumor location(recum or colon),depth of invasion and tumor differentiation,there was no significant difference (P>0.05).COX-2 protein expression in lymphnode metastasis positive group compared with lymphnode metastasis negative group,there was significant difference between the groups(P<0.05). COX-2 protein expression in Dukers A/B group was 61.53%, in Dukers C/D group was 86.95%, there was significant difference between the groups (P<0.01).5. There was 81 cases with positive expression of VEGF protein in 98 cases of colorectal carcinoma, negative expression was 17 cases, There was 72 cases with positive expression of COX-2 protein in 98 cases of colorectal carcinoma, negative expression was 26 cases, Which VEGF positive/COX-2 positive was 58 cases, VEGF negative/COX-2 positive was 14 cases, VEGF positive/COX-2 negative was 23 cases, VEGF negative/COX-2 negative was 3 cases. Under the spearman correlation analysis, VEGF protein and COX-2 protein expression in colorectal cancer tissues showed a significant positive correlation (rs=0.367, P<0.01).6.MVD in cases with VEGF-positive colorectal carcinoma was 39.83±7.97, MVD in cases with VEGF-negative colorectal carcinoma was 23.78±8.56, there was significant difference between the groups(P<0.01).7. MVD in cases with COX-2-positive colorectal carcinoma was 39.88±9.33, MVD in cases with COX-2-negative colorectal carcinoma was 29.19±7.77, there was significant difference between the groups(P<0.01).Conclusion:1. Colorectal carcinomas showed high expression of VEGF protein, which shows a close correlation with histological grade, invasion, lymphrode metastasis, Dukers' stage and angiogenesis, which suggests that VEGF expression is closely related to colorectal carcinogenesis and development.2. The MVD labeled with CD34 in colorectal carcinoma was significantly higher than that of adenomas and normal mucosa samples, which suggests that colorectal carcinoma angiogenesis is closely related to tumor growth.3. Colorectal carcinomas showed high expression of COX-2 protein, which shows a close correlation with lymphrode metastasis, Dukers'stage and angiogenesis, which suggests that COX-2 expression is closely related to colorectal carcinogenesis and development.5. There exists a positive correlation of the positive expression between COX-2 and VEGF, which suggests COX-2 may be an important factor in inducing the up-regulation of VEGF.
