bjective:To investigate the effects and possible mechanism of Peripheral blood Mesenchyme Stem CelIs(PBMSCs) from bone marrow cells mobilization translation on restenosis in Atherosclerosis model rabbits carotid postangioplasty.Methods:Bone marrow cells mobilization by granulocyte colony-stimulating factor(G-CSF)30μg/(kg·d) subcutaneously for 5 days,The sixth day gather the Peripheral blood);To obtain purified PBMSCs by density gradient centrifugation and adherent culture then identificated it by flow cytometer.Amplificating it in vitro,keep a great quantity of them by freezing.and labeled it by enhancement type green fluorescent protein(GFP) reporter genes before translated,rabbits undergoing carotid Atherosclerostenosis were randomly divided into two groups transplanted group(T group,N =30) was received intravenous transplantation of PBMSCs and control group(C group,N=30) was only received intravenous equal-volume culture solutions following right common carotid balloon angioplasty.The animals were killed 7,14,28days(n=10) after operation respectively and then took the vessels tissues and blood specimen for survey and evaluation.Help with image analysis system and morphometric analysis of injuried carotid artery neointima formation by Weigert elastic fibers staining;immunohistochemisty analysis of the transplanted cells homing,differentiation and injuried carotid artery reendothelialization,expression of GFP,apoptosis of smooth muscle cells;levels of vascular endothelial growth factor(VEGF) and C reactive protein(CRP) in blood were assayed by ELISA;The changes of liver,renal function and creatase were detection before and after operation.Results:①MSCs could be separated from Peripheral blood and amplified in vitro after bone marrow cells mobilization by G-CSF②MSCs were good for labeled by infection of adenovirus vector with GFP gene③Levels of VEGF in serum was higher of T groups than C groups and the tunica media thickness and restenosis ratio lower obviously but the areas of endothelialization increase in T groups,compared with C groups.④Parts of endotheliocytes were GFP positive in T groups.⑤The apoptosis ratio of smooth muscle cells in injuried vessels local significant was higher in T groups compared with C groups.⑥Levels of CRP,liver,renal function and creatase in serum was no significant deviation in both group.Conclusion:Intravenous transplantation of PBMSCs accelerate reendothelialization and attenuate neointimal thickening and restenosis in rabbits carotid balloon-induced injury model.The beneficial effects mayby ascribed into MSCs secretion of VEGF and homing to the injured vessel local then differentiating into endotheliocytes.And there is no causing of phlegmonosis and myocardial damage to laboratory animals in the near future. |
