Expression And Significance Of Smac And Survivin In Cervical Squamous Carcinima | | Posted on:2010-04-23 | Degree:Master | Type:Thesis | | Country:China | Candidate:C M Yan | Full Text:PDF | | GTID:2144360302958449 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | OBJECTIVE To investigate the expressions of second mitochondria-derived activator of caspase and Survivin proteins in Cervical Squamous Carcinoma and explore their clinical significance.METHODS The expressions of Smac and survivin proteins in 70 cervical squamous carcinoma tissues and 11 normal cervical tissues have been detected using strep tavidin-bintin peroxidase (SP) method,and their correlations with cervical squamous carcinoma have been analized.RESULTSSmac proteins were expressed in 35 of 70 (50%) cases of cervical squamous carcinomas.However,it was not expressed in normal cervical tissues. Positive expression rate of Smac proteins are 80.95%, 50.00% and 19.04% in clinic stage I, stage II and stage III in cervical squamous carcinoma, respectively(P<0.05). There was no relationships between the expression of Smac proteins with tumor grade or lymph node metastases. Survivin proteins were expressed in 38 of 70(54.29%) cases of cervical squamous carcinomas , but not expressed in normal cervical cases. The positive expression rate of Survivin proteins in cervical squamous carcinoma was 27.27%(3/11), 59.32%(35/59) in grade G1 and G2 , G3, respectively (P<0.05). 52.5% of Survivin proteins expression was correlated with clinic stage I and II,and 80.96% of Survivin proteins expression was correlated with clinic stage III (P<0.05). Positive expression of survivin proteins was positive related to the tumor grade and clinical stage (r=0.328,r=0.345,P<0.05). The expression level of smac and survivin showed negative correlation (r=-0.236,P=0.016).CONCLUSIONS1.The positive expression rate of Smac and survivin proteins were up-regulated in cervical squamous carcinoma. Smac and Survivin proteins may play important roles in oncogenesis and progression of cervical carcinoma.2. The expression of Smac gene is a frequent event in cervical carcinogenesis.Smac promotes apoptosis of cervical cancinoma cells by suppression of Survivin.
