| 1.ObjectiveWe detect the expression of peripheral blood interleukin-7(IL7) of HIV-1 infected and the impact of long-term HAART,simulate HIV-1 infection and IL7 effect on the cell proliferation and activity of lymphocytes in vitro,investigate the impact of HIV-1 infection to IL7 and its receptor system.If HIV-1 infection has a significant effect on the expression of IL7,then what about long-term HAART's effect?Besides,HIV-1 is used to infect human lymphocytes in vitro in order to study IL7's effect on T lymphocyte cell proliferation and activity in vitro,as well as the immune relationship with HIV-1 infection,which are summed up to discuss IL7's role in immune reconstruction and its change mechanism induced by HIV-1 infection2.Material and MethodsWe use 28 cases of HIV-1 infected from Youth hospital,China,and 92 cases from German Ruhr Center,test CD4,CD8,NK,CD38,HLADR cells,IL7 protein concentration and HIV-1 viral load of the patients' peripheral blood.Meanwhile pcDNA3.1(+)-IL7-293T cell lines in vitro with stable expression of IL7 protein were reconstructed,the cell proliferation and activity of human MT2 cells under IL7 protein and HIV-1 were observed. 3.Results1,CD4,NK cell of HIV-1 infected decreased significantly(average p<0.05),CD8 cells,HLADR,CD38,and IL7 were significantly higher than normal(average p<0.05).2,HAART can significantly increase CD4,NK(average p<0.05),decrease HLADR, IL7 levels(average p<0.05),but still can't reach the normal level(p<0.05 ).CD38 goes down slowly during HAART.3,IL7 protein in vitro alone could significantly promote the cell proliferation and activity of MT2 cells(p<0.05),which will decrease to some degree after HIV-1 effect(p<0.05),while still higher than the normal group(p<0.05)4.Conclusion1.HIV-1 infection can damage large number of immune cells,promote the expression of immune activation indicators and IL72.HAART can effectively inhibit HIV-1 viral replication,prevent the destruction of immune cells and reduce the body's abnormal immune activation levels to a certain extent,which still can't be recovered completely.During HAART process,IL7 level decreases gruadually,which also can't be recovered to normal,suggesting IL7 system may play an important role in HIV-1 infection and immune reconstitution.3.pcDNA3.1(+)-IL7-293T cell line,with a stable expression of IL7 protein,is reconstructed successfully.4.IL7 can significantly promote the cell proliferationo and activity of lymphocytes.Besides direct destruction to lymphocytes,HIV-1 possibly damage IL7 and its receptor system function through other ways. |
