The safety of milk and dairy products became the world's focus of attention with the melamine incident in dairy products. To overcome the original milk protein detection method with low specificity, the establishment of milk protein detection methods with strong specificity and great efficiency is of great significance.The polyclonal antibodies were prepared by immunizing mice with bovine serum albumin and casein separately in this paper. Enzyme-Linked Immunosorbent Assays (ELISA) was developed for bovine serum albumin and casein separately and the specificity of the assays was tested. The establishment of milk protein detection methods with milk-based protein ELISA and the correlation between the ELISA methods were explored.The experimental results indicated that the casein accounted for 43.56% and bovine serum albumin accounted for 4.55%. The bovine serum albumin accounted for 83.04%and bovine serum albumin accounted for 29.98%. The differences in total protein content and the two kinds of specific proteins were great in yogurt. So the total protein in milk and milk powder can be detected with casein and bovine serum albumin.The protein detection methods for milk with strong specificity and efficiency were initially established suggesting potential efficacy in the protein detection for milk with ELISA.
