Study On The Association Between Epstein-Barr Virus-encoding LMP-1 With Ets-1 And Cyclin D1 Expression In Nasopharyngeal Carcinoma

Objective:Many studies indicated that Latent membrane protein 1 (LMP-1) encoded by Epstein-Barr virus (EBV) played a key role in nasopharyngeal carcinoma (NPC), However, the underlying molecular mechanism of NPC carcinogenesis is still unclearly known. Our previous studies found that LMP-1 expression was associated with Cyclin D1. To be most concerned, LMP-1 can activate the Ras signaling pathway via the Ras/MAPKKK/MAPKK/ERK/Ets-1 signal, that is LMP-1 can regulate cell signal pathway by activating Ets-1. Furthermore, a study suggested, Ets-1 has a combining Site (5'-GATCTCGAGCAGGAAGTTCGA-3') on the cyclin D1 promoter region in mouse renal cell, and could active the promoter, increase cyclin D1 expression. Therefore, we supposed that there was a correlation of expression between LMP-1, Ets-1 and Cyclin D1 in NPC. The aim of this study was to study the expression of LMP-1, Ets-1 and Cyclin D1, and the association between them, which can provide evidence for further study of LMP-1 in carcinogenesis of NPC.Methods:We used immunohistochemistry (Envision method) to detect the expression of LMP-1,Ets-1 and Cyclin D1 in nasopharyngeal carcinoma (nonkeratinizing squamous-celled carcinoma) and analyze the association between them. Among them, male patients 96, female patients 47, the average age was 48.2±14.8, and the age range from 16 to 82.Results:●Expression of LMP-1,Ets-1 and Cyclin Dl in NPC In 143 nasopharyngeal carcinoma cases, the positive percentage of LMP-1 expression was 81.8%(117/143); Ets-1 was positively expressed in 95.1% (136/143) of the cases; and cyclin D1 expression was positive in 91.6% (136/143) of the cases. All of them were high expression in NPC.●The association between LMP-1, Ets-1, and Cyclin D1 expression The result of correlation analysis showed that high expression of LMP-1 was significantly associated with Ets-1 and Cyclin D1 (LMP-1 Vs Ets-1, P = 0.000, r = 0.397; LMP-1 Vs Cyclin D1 P= 0.001, r= 0.315), and we also found that Ets-1 was associated with Cyclin D1 expression (P= 0.004, r= 0.368).Conclusions:The expression of LMP-1, Ets-1 and Cyclin D1 are high in NPC, and have a significantly correlation between them.
