| Objective: To research the expression of pl4ARF protein in breast carcinomas tissue.To investigate their relationships of the expression of p14ARF protein in breast carcinomas tissue and adjacent noncancerous breast tissue,hyperplasia of mammary glands tissue,common breast tissue,and to analyze their relationships of the expression of p14ARF protein in breast carcinomas tissue with the age of patients,tumor -size,tissue-classify,clinical-stage,state of estrogen and progesterone receptor and axillary lymph node transferred ,and to expect to provide the experimental basis of diagnosis,gene therapy and prognosis judgement for breast carcinomas.Methods: All tissues to draw the materials from were fixed by 10% formol and paraffin embedding and paraffme specimen serial section . The specimens were ofen staininged by hematoxylin-feosin. The expression of p14ARF protein were studied in 56 cases of breast carcinomas tissue,20 cases of adjacent noncancerous breast tissue,20 cases of hyperplasia of mammary gland tissue,20 cases of common breast tissue by immumohistochemical technology(S-P). Their relationships of the expression of p14ARF protein in breast carcinomas tissue,adjacent noncancerous breast tissue,hyperplasia of mammary gland tissue and common breast tissue and the correlation between the expresson and clinical-pathologic parameters wereanalyzed.Result: The immunostaining of p14ARF protein is localized mainly in nucleus of breast carcinomas tissue,adjacent noncancerous breast tissue,hyperplasia of mammary glands tissue and common breast tissue. The positive expression rate of p14ARF protein was 46.4%(26/56) in breast carcinomas tissue, and significantly lower than that in adjacent noncancerous breast tissue85.0%(17/20), innocence mammary glands tissue90.0% ( 18/20 ) and common breast tissue100.0% (20/20)(p<0.0083),and there were no statistics distinction among the rate in adjacent noncancerous breast tissue,hyperplasia of mammary gland tissue and common breast tissue (P>0.0083) . The positive expression rate of p14ARF protein in poorly differentiated breast carcinomas tissue,moderately differentiated breast carcinomas tissue and well differentiated breast carcinomas tissue were 22.2%(4/18),45.5%(10/22) and 75.0%(12/16),respectively.The positive expression rate of p14ARF protein was significantly lower in poorly differentiated breast cnacer tissue than well-differentiated . There was statistics distinction among them (P < 0.016 ) , and there were no statistics distinction between moderately differentiated breast carcinomas tissue and well differentiated and between moderately differentiated and poorly differentiated (P>0.016).The positive expression rate of p14ARF protein was significantly lower in breast carcinomas tissue with axillary lymph node metastasis (29.0%) than that in breast carcinomas tissue without axillary lymph node metastasis (68.0%) (p<0.05),and it did not correlate with age, tumor size, TNM staging, ER and PR.et al(P>0.05).Conclusions: 1,There is down-regulation of the expression of p14ARF protein in common breast tissue,innocence breast tissue,adjacent noncancerous breast tissue and breast carcinoma tissue. The positive expression rate of pl4ARF protein was in breast carcinomas tissue significantly lower than that in adjacent noncancerous breast tissue, innocence mammary glands tissueand common breast tissue,and there were no statistics distinction among the rate in adjacent noncancerous breast tissue,hyperplasia of mammary gland tissue and common breast tissue. 2,The level of the expression of p14ARF protein was significantly lower in poorly differentiated breast cnacer tissue than well-differentiated, and there were no statistics distinction between moderately differentiated breast carcinomas tissue well differentiated and between moderately differentiated and poorly differentiated.The level of the expression p14ARF protein in breast cancer with lymph node involvement was significantly lower than in breast cancer without lymph node involvement, and it did not correlate with age, tumor size, TNM staging, ER and PR.et al. 3,These observations define that p14ARF is a suppressor gene. down-regulation of p14ARF may be implicated in the carcinogenesis and invasion and metastasis of breast carcinoma. |
