Objective To observe ultrastructural morphological changes of pathogenic yeast form of Penicillium marneffei prasited in Guangxi wild Rhizomys pruinosus and clinical isolates before and after treated by Amphotericin B and Voriconazole,and explore the relevance of different originated of PM and provide a more intuitive basis to the clinical drug of PSM.Method After determinated the minimum inhibitory concentration (MIC)of Amphotericin B and Voriconazole to yeast form of Penicillium marneffei prasited in Guangxi wild Rhizomys pruinosus and clinical isolates by microdilution broth method,We used the drugs concentration of MIC and 10MIC to treat the different originated of PM in their logarithmic phase of the bacteria in 37℃.After 24,48 and 72h of incubation in 37℃,all the cultures were harvested and examined by the scanning electron microscopy(SEM) and transmission electron microscope(TEM) .Result With the exposure time and concentrations,thallus of yeast form of PM parasitic in Guangxi wild Rhizomys pruinosus and clinical isolate treated by Amphotericin B were all broken, peeled in the outer layer, shrinkaged, complete broken, fractured and adhesion ultimately by SEM; In the ultrathin,we could observe the degenerated mitochondria,fragmental nucleus,shrinkage of cytoplasmic membrane,complete fragmentation of intracellular organelles, fracture of cell wall at last by TEM.After treated by Voriconazole,the thallus of different originated of PM were all damaged,wrinkled,shriveled and collapsed ultimately by SEM; In the ultrathin,there were many high-density electron-dense granules in cytoplasm,degenerated mitochondria,rough cell wall in the surface,shrinkage and damage of cytoplasmic membrane,degeneration of intracellular organelles finally were observed by TEM.Conclusion Ultrastructure changes of yeast form of the two different originated of PM after treated by Amphotericin B and Voriconazole are have severe damage,but the changes is not significant difference to the two different originated of PM.These results also suggest that the effect of two kinds of antifungal drug are similar to the two different originated PM.
