| Objeetive: To explore the renlationship between 3-nitrotyrosine and cardiac cell apoptosis of diabetic cardiomyopathy in rat.Methods: SD rat(male, 8W) were randomly divided into four groups of 15 animals each: the control group(N group); the diabetic cardiomyopathy group(D group); the diabetic group pretreatmented with valsartan(40mg/kg/d) (D+P group); the DCM group treatmented with valsartan(40mg/kg/d) (D+S group). Apoptotic index(AI) of cardiac myocyte of diabetic cardiomyopathy in rat was examined by TUNEL. Expression index(EI) of 3-nitrotyrosine in rat cardiac myocyte was examined by immunohistochemisty.The 3-nitrotyrosine concentration of blood serum in rat was examined by ELISA.Results: (1)The cardiac weight index comparison of four group had statistical significance(P<0.01). Compared with the N group and D+P group the cardiac weight index was higher in the D group and D+S group(P<0.05). (2) In each group AI and EI had direct correlation(allP<0.01). But AI and the 3-nitrotyrosine concentration of rat blood serum in each group had no correlation(all P>0.05). (3) The four group comparison of AI had statistical significanc(eP<0.01). AI in the D group was highest than other group(P<0.05). And compared with the N group AI was higher in the D group and D+S group(P <0.05). (4) The four group comparison of EI had statistical significance(P<0.01). Compared with other group EI in the D group was highest(P<0.05). (5) The four group comparison of 3-nitrotyrosine concentration in rat blood serum had no statistical significance(P>0.05).Conclusions: (1) DCM myocardial tissue 3-NT expression is significantly increased with the closely related cardiac cell apoptosis, valsartan can inhibit DCM myocardial cells 3-NT expression, and thus inhibit the DCM myocardium apoptosis. (2) The circulating blood of 3-NT levels can not be a true reflection of DCM myocardial tissue 3-NT expression and its effect on myocardial cell apoptosis. (3) Notwithstanding Valsartan inhibites 3-NT-induced cardiomyocyte apoptosis in the myocardial tissue, but no significant improvement of enlargement of the heart, indicating that apoptosis of myocardial cells is not the only diabetic cardiomyopathy rat heart expanding pathogenesis. |
