| ObjectiveIdiopathic pulmonary fibrosis is an unexplained,choronic fibrosing interstitial Pneumonia , The pathology change take becomes the fibroblasts, the extracellular matrix excessively deposition as a characteristic . In recent years the disease incidence rate assumed year by year the above tendency,on clinical lacks the effective treatment method. The traditional anti-inflammatory, immunosuppressants curative effect is not ideal, side effects. As the pathogenesis of this disease continue to in-depth study of the treatment, also from the traditional anti-inflammatory changes to the anti-fibrosis therapy. Human embryonic lung fibroblasts in vitro may reflect the status of pulmonary fibrosis. Therefore, the purpose of this study human embryonic lung fibroblast cells, observation of chloral hydrate to its inhibition of proliferation and cell apoptosis. Discussion chloral hydrate inhibition of pulmonary fibrosis,provides the experiment and the theory basis.Methods(1)Detected the influence of CH on HELF cells growth at different concentrations and time points by MTT. (2)Detected the influence of CH on apoptosis and cell cycle of HELF cells by FCM.(3)Observed under transmission electron microscope human embryonic lung fibroblasts microscopic structural changes.Results(1)CH can inhibit HELF cell growth ,and with time-effect and the dose-effect dependence.(2)CH can induce apoptosis of HELF cell, and has a dose-effect dependence.(3)CH can make cell lines HELF arrest at G1 phase in a dose-effect dependence. Conclusion Chloral hydrate on human embryonic lung fibroblast proliferation and inhibit induction of apoptosis.(4)Transmission electricity observation CH acts on the HELF cell down under the mirror:Cell shrinkage,irregular nature of the nuclear,nuclear matrix, dense cytoplasm, the formation of vacuoles. |
