Objective:Migraine is a commonly primary headache,the pathogenesis is not completely described so far. That is a disease we often discover it towards the female. It begins in adolescence, happens in premenstrual period or menstrual period, relieves or disappears in menopause, stops in pregnancy, and recurrences after obstetric. So suggest the unique female menstrual period physiological variation when migraine take place. The reason of estrogen level steeped down has reached a consensus generally. But how it takes part in migraineis is still mysterious.It's SP that belongs to the peptides. Now we consider that, SP takes dual-action in paining experience: to the external to transmit messages to nervus centralis---this can facilitate the delivery of paining experience; and to the nervus centralis system it mianly play a major function as analgesia. The function is indivisible with the PAG. Estrogen may participate in migraine pathogenesis by the way of SP,as a lot of research indicates that,but we don't know how at present.In our medical research, we make Nitroglycerinizm migrainerats models. In order to eliminate the effective of diverse estrogen level, we took the ovaries of female rats of. And instead we supplied peanut oil,low-dose and high-dose estrogen to held the estrogen level. By the observation of behavioristics change of migrainerats to discuss the function of estrogen to migraine. And we also investigate migraine rats's SP of its PAG and the effection of receptor NK1 expression. This is of the purpose to discuss the function and pathogenesis of estrogen prevention and treatment migraine,and to provide basis on prophylaxis and therapy.Method:①Take a row of 36 female Wistars ovariectomized surgery.②The rats were randomly divided into six groups: comparison group, migraine group, low-dose estrogen cure group, high-dose estrogen cure group, low-dose estrogen comparison group, high-dose estrogen comparison group, 6 rates each group.③For 7 days after ovariectomy, the comparison group, migraine group were given intraperitoneal injection of 0.1ml peanut oil, low-dose estrogen cure group, low-dose estrogen comparison group were given 0.1mg/kg intraperitoneal injection of estradiol, high-dose estrogen cure group and high-dose estrogen comparison group, were given 1.0 mg / kg intraperitoneal injection of estradiol. By the 15th day after surgery, the migraine group,low-dose estrogen cure group,high-dose estrogen cure group, were given 10mg/kg subcutaneous nitroglycerin to make preparation of migraine models, comparison group,low-dose estrogen comparison group,high-dose estrogen comparison group were given peanut oil.④Take observations of behavioristics change on rats.⑤Take the midbrain, immunohistochemical staining to detect the expression of SP and its receptor NK1.Results:①Behavioristics change:compare with comparison group,low-dose estrogen comparison group,high-dose estrogen comparison group,we found migraine group,low-dose estrogen cure group,high-dose estrogen cure group present significant difference (P<0.05).Compare with high-dose estrogen cure group ,migraine group and low-dose estrogen cure group present significant difference (P<0.05). Compare with high-dose estrogen cure group , low-dose estrogen cure group present significant difference (P<0.05).②Count midbrain SP immunoreactivity neuron of PAG:The rates compare with comparison group ,migraine group PAG by midbrain trochlear nucleus level SP immunoreactivity neuron present no significant difference(P>0.05).low-dose estrogen cure group,high-dose estrogen cure group,low-dose estrogen comparison group,high-dose estrogen comparison group PAG by midbrain trochlear nucleus level SP immunoreactivity neuronobviously higher than migraine group and comparison group (P<0.05). low-dose estrogen cure group,high-dose estrogen cure group,low-dose estrogen comparison group,high-dose estrogen comparison group PAG by midbrain trochlear nucleus level SP immunoreactivity neuron present no significant difference(P>0.05).③Count midbrain PAG's SP receptor NK1 immunoreactivity neuron: each group of rats: comparison group,migraine group,low-dose estrogen cure group,high-dose estrogen cure group,low-dose estrogen comparison group,high-dose estrogen comparison group midbrain PAG trochlear nucleus level NK1 receptor immunoreactivity neuron present no significant difference(P>0.05).Conclusion:①It's estrogent hat can mitigate migrainerats behavioristics changes,the curative effect depends on it also.②estrogenmay increase the expression of SP of PAG migrainerats,prompt it's function of prevention and the treatment migraine function in relation to the expression of SP of PAG。 ③Estrogen has no obviously effective to the expression of PAG's SP receptor NK1.It is still need more study that whether estrogenhas regulation to the expression of nervus centralis system SP receptor NK1,and so as it's mechanism.
