| IntroductionCell growth environment includes the extracellular matrix and adjacent cells. The biological function of extracellular matrix contain affecting cell growth and cell death,determining the shape of cells,controlling cell differentiation and involving cell migration.These important biological functions are completed mainly by cell adhesion and extracellular matrix. Migfilin is a cytoskeletal protein, and is widely distributed in cell-cell and cell-matrix adhesion. FBLIM1 gene which is located in 1p36 encode Migfilin. And its molecular weight is 46KD.Migfilin contains an N-terminal region that interacts with Filamin, a central proline-rich region and a C-terminal region consisting of three LIM domains. LIM domains are cysteine-rich sequences of about 50 residues that fold into a specific two-zinc-finger structure.But we do not know if Migfilin is involved in CyclinB1-induced invasion and metastasis of esophageal squamous carcinoma cell lines.This paper discusses the relationship between Migfilin and CyclinBl-induced invasion and metastasis of esophageal squamous carcinoma cell lines. The researchers found CylinB1 can induce NF-kB dependent EMT, which cause the invasion and metastasis of esophageal squamous carcinoma cell lines.Migfilin as a cytoskeletal protein may play an important role in that process.MethodsFirstly,the mRNA expression of Migfilin is detected by RT-PCR.Secondly, the protein expression of Migfilin is detected by Western Blot.Thirdly,we detect the subcellular location of both cell lines.Fourth,Migfilin is knocked down by RNAi in high-cycB1 esophageal squamous carcinoma cell lines.Finally,After knocking down Migfilin successfully,we detect invasion ability in high-cycB1 esophageal squamous carcinoma cell lines by Transwell.And we also detect the ability of single cells into tumor in high-cycB1 esophageal squamous carcinoma cell lines by colony formation. ResultsMigfilin and CyclinB1 did not show significant correlation in mRNA level.However, expression of both show positive correlation in protein level. The expression of Migfilin varies as same as CyclinBl.With the reduction of Migfilin, the ability of invasion and colony formation will reduce in high-cycB1 esophageal squamous carcinoma cell lines.ConclusionThe expression of Migfilin and CyclinB1 show positive correlation in high-cycB1 and CycBl-siRNA esophageal squamous carcinoma cell lines. And.immunofluorescence results show the phenomenon of colocalization.After knocking down Migfilin,invasion ability is reduced in in high-cycB1 esophageal squamous carcinoma cell line without changing the expression of CyclinB1.With the reduction of Migfilin,colony formation will in high-cycB1 esophageal squamous carcinoma cell lines.So,Migfilin plays an important role in the CyclinB1-induced invasion and metastasis of esophageal squamous carcinoma cell lines. |
