Objective:To study the effects and the mechanisms of Liangxue Jiedu Huoxue Granule on the prevention and treatment of radiation pneumonitis.Methods:210 wistar male rats were randomly divided into blank group(A group), model group(B group), positive medicine group(C group), low dose treatment group(D group), middle dose treatment group(E group) and high dose treatment group(F group). The rats in A group received sham irradiation; others were irradiated with a single dose of 25 Gy to their right hemi thoraxes by a 6MVX-ray. The rats in A and B groups were given saline a week before irradiation, while rats in D, E and F groups were given different dose of Liangxue Jiedu Huoxue Granule at the same time. Rats in C groups were given prednisone after irradiation. All drugs were administered by intragastric administration (i.g) once a day. At the time of 7d,30d and 90d after irradiation, the effects of drugs were evaluated according to pathological changes, the right lung index, the levels of IL-6 and TGF-β1 from the lung homogenate; And the mechanisms were also analyzed according to the levels of ACTH and Cor, the changes of whole blood viscosity and coagulation indexes, the blood routine examination, the index of thymus gland, adrenal glands and spleen. Models of acute inflammatory reaction were studied by acetic acid-induced elevation of celiac capillary's permeability and egg white-induced paw edema.Results:1. The rats in B group developed typical radiation pneumonitis and fibrosis. And the levels of IL-6 and TGF-β1 had increased after irradiation. As compared with B group, the pathological changes of radiation injury were slighter and the levels of IL-6 and TGF-P1 had decreased in C~F groups. 2. Prednisone and Liangxue Jiedu Huoxue Granule could suppress acetic acid-induced elevation of celiac capillary permeability and egg white-induced paw edema.3. The rats in B group were in the high coagulation and viscosity state in 7d, in the low coagulation and viscosity state in 30d, in the low coagulation and high viscosity state in 90d. The coagulation time in the D~F group was longer in 7d, shorter in 30d and 90d; The whole blood viscosity had decreased in 7d, increased in 30d, decreased in 90d.4. ACTH in B group had decreased in 7d, then increased in 30d and 90d; and Cor had no change at first, and then decreased. As compared with B group, ACTH and Cor had increased in E group and F group.5. The index of adrenal glands in B group had decreased in 90d. The index of adrenal glands in C group had increased in 30d, decreased in 90d. And that had increased in C~F groups than that in B group.6. The number of white blood cells in B group had significantly decreased after irradiated. The index of thymus gland and spleen in B group had decreased after irradiation. The index of thymus gland in C group had decreased. As compared with B group, the number of white blood cells in D-F groups had increased; And the index of thymus gland and spleen had also increased.Conclusion:1. The rat model of radiation pulmonary injury can be successfully established by a single dose of 25Gy irradiation to the right hemi thorax.2. Liangxue Jiedu Huoxue Granule can ameliorate symptoms of radiation pulmonary injury, prevent radiation pneumonitis, and relieve the pulmonary fibrosis. The high dose can get the best result.3. There are two main mechanisms of Liangxue Jiedu Huoxue Granule on the prevention and treatment of radiation pneumonitis.(1) The herbal granule plays anti-inflammatory role mainly by the regulation of the hypothalamus-pituitary-adrenal axis (HPA axis). (2) The herbal granule can make appropriate adjustment of blood coagulation function disorder and viscosity abnormality occurred in the radiation-induced lung injury.4. Liangxue Jiedu Huoxue Granule can also improve the body's immunity, and slow down the decline of white blood cells after radiation...
