The Effect Of Shen Shuai Fang On Expression Of VEGF And TSP-1 In Renal Tissue Of Renal Interstitial Fibrosis Rat Model

Objective:To Study the micro mechanism of Shenshuaifang Granules on treating renal interstitial fibrosis in chronic renal failure by observing the functional and morphological changes of the kidney and detecting the amounts of VEGF, TSP-1 and TGF-β1 in renal tissues of rats.Methods:60 male SD rats were randomly divided into normal group(group A,8 rats) and experimental group(52 rats). Rats of the experimental group were gavaged with adenine for twenty four to induced renal interstitial fibrosis. The successful models were randomly divided into model group(group B), Shenshuaifang Granules high dose group(group C), Shenshuaifang Granules low dose group(group D), losartan group(group E) and high dose Shenshuaifang Granules with losartan group(group F). Every group were given the relevant medicine for 4 weeks. During the experiment, weights of all rats were recorded every week and SCr, BUN,24h urinary protein were detected. Pathological changes of the kidney were ovserved by hematoxylin and eosin staining of pathological sections. VEGF, TSP-1 and TGF-β1 in renal tissue of rats were detected with immunohistochemical assay.Results:Scr, BUN,24h urinary protein, body weight, kidney weight and kidney index of group C, group D and group F all decreased significantly (P< 0.01),compared with group B, and indexes of group C and group F decreased most obviously. Compared with group B, VEGF of group C and group F increased significantly (P< 0.01), TSP-1 of group C, group D, group E and group F decreased significantly (P<0.01). Pathological damage of group C, group D, group E and group F were milder than group B, and group C and group F were the mildest.Conclusion:Shenshuaifang Granules reduced Scr, BUN and 24h urinary protein, and slowed down the progress of renal fibrosis. Regulating expressions of VEGF, TSP-1 and TGF-(31 in the kidney tissue may be its effective mechanism.
