OBJECTIVE:To study expression, correlations and significance of P21ras,PIK3CA and P-AKT in Hepatitis B virus-related hepatocellular carcinoma, Post-Hepatitis B hepatic cirrhosis and normal hepatic tissue. Our study intend to explore function and significance of P21ras,PIK3CA and P-AKT in Hepatocellular carcinoma and in the course of tumorigenesis that develop from Post-Hepatitis B hepatic cirrhosis to Hepatitis B virus-related hepatocellular carcinoma through detect the expression of P21ras,PIK3CA and P-AKT protein with the method of immunohistochemistry in Post-Hepatitis B hepatic cirrhosis and Hepatitis B virus-related hepatocellular carcinoma tissue specimen.METHODS:34 Hepatitis B virus-related hepatocellular carcinoma,37 Post-Hepatitis B hepatic cirrhosis and 30 normal hepatic tissue specimen were collected from the Department of second General Surgery in the First Hospital of Lanzhou University between August 2008 and November 2006. The tissues were stored at -80℃. All the cases were confirmed by operation and pathology, and all the cases were not experienced radiotherapy and chemotherapy before operation. Detection the expression of P21ras,PIK3CA and P-AKT in all specimens by immunohistochemistry. Using SPSS 13.0 software. Probability values less than 0.05 were considered to indicate statistical significance.RESULTS:(1) The mean gray scale of P21ras protein in Hepatitis B virus-related hepatocellular carcinoma, Post-Hepatitis B hepatic cirrhosis and normal hepatic tissue specimen were 138.86±2.9,145.34±2.06 and 152.07±1.17 (P<0.01) respectively, and it is related to the progression of hepatopathy(P<0.01). (2) The mean gray scale of PIK3CA in Hepatitis B virus-related hepatocellular carcinoma (138.20±3.14) was higher than in the Post-Hepatitis B hepatic cirrhosis (149.49±0.78) and normal hepatic tissue (154.71±1.29), there was a significant deviation among different progression of hepatopathy (P<0.01). (3) The expression of P-AKT was similar to PIK3CA in three groups, its expression was Hepatitis B virus-related hepatocellular carcinoma (146.83±3.20), Post-Hepatitis B hepatic cirrhosis (150.97±2.59) and in normal hepatic tissue (155.00±4.37) respectively, and there was a significant deviation among different progression of hepatopathy (P<0.01). (4)There was a apparent correlation between P21ras and PIK3CA protein (r=0.64, P=0.020), PIK3CA and P-AKT(r=0.594, p=0.001), but no correlation was found between P21ras and P-AKT protein (r=0.145, P=0.315) in Hepatitis B virus-related hepatocellular carcinoma and Post-Hepatitis B hepatic cirrhosis (r=0.48, p=0.02) respectively. It was similar to Hepatitis B virus-related hepatocellular carcinoma tissues, there was a apparent correlation between P21ras and PIK3CA protein (r=0.547, P=0.001), PIK3CA and P-AKT(r =0.378, p=0.001), but no correlation was found between P21ras and P-AKT protein (r=0.141, P=0.334) in Post-Hepatitis B hepatic cirrhosis group.CONCLUSION:The expression of P21ras,PIK3CA and P-AKT in Hepatitis B virus-related hepatocellular carcinoma tissues were more higher than in Post-Hepatitis B hepatic cirrhosis and normal hepatic tissue, this three factor interact one another, and participate in the occurrence, development of Hepatitis B virus-related hepatocellular carcinoma in common. Probably, P21ras-PIK3CA-P-AKT signal pathway play a important role in the course of tumorigenesis that develop from Post-Hepatitis B hepatic cirrhosis to Hepatitis B virus-related hepatocellular carcinoma, as a result to promotes hepatocyte proliferation, suppresses apoptosis and induces hepatocellular carcinoma.
