The EC(endometrial cancer) is a malignant neoplasm which is originated from endometrial epithelial.In America,Its incidence rate is the first in gynaecological cancer. It accounts 7% female cancer and 20%-30% gynaecological genital cancer.Traditional therapy are urgery, radiotherapy, chemotherapy and hormone therapy.However, the five-year survival rate is only 60%-70%.Therefore, searching for new therapeutic methods is become to one of the focal point of the cancer research. For the past few years, molecule target therapy and the traditional Chinese medicine therapy are gradually focus researcher's attention upon cancer.This also indicates a new method to endometrial cancinoma therapy.20(s)-ginsenoside Rg3 is Chinese herbal drugs monomer micro-selected by the Red gindeng.During the past decade,many researchs confirmed that Rg3 have medicinal effects,such as anti-inflammatory, anti-aging activities and anti-tumorigenic. The anti-tumorigenic effect on lung, gastric, melanoma, pancreatic, colorectal, liver,breast etal tumors have been reported. The growth and proliferation of tumorous are closed related with cell apoptosis. consequently, it is a important means to induce tumor cell apoptosis through the traditional Chinese medicine.LewisY antigen is a tumor-related carbohydrate antigen and is over-expressed on the surface of human epithelial cancers including sixty to ninety percent breast cancer, colorectal Cancer, ovarian cancer, prostate cancer, pancreatic cancer and non-small-cell lung carcinoma. The current research indicates Lewis Y antigen is not expressed in normal uterine endometrium,however,it is over-expressed in simple and complex endometrial hyperplasia, atypical hyperplasia and endometrial carcinoma and is mainly expressed in cytoplasm and cytomembrane of glandular epithelium.It is not reported about targeted-LewisY antigen antibody therapy on endometrial cancer in home and abroad.We used the LewisY-specific antibody combining 20(s)-ginsenoside(Rg3) to demonstrate the inhibitory effect on endometrial cancinoma cell growth in vitro and to explore the mechanism through the expression of Bcl-2 and Bax.This study aimed to establish experimental evidence for the biotherapy of endometrial cancinoma.Objective:To investgate inhibitory effects of combining 20(s)-ginsenoside Rg3 and anti-LewisY antibody on endometrial carcinoma cellline A431 growth and expression of bcl-2 and bax protein in vitro.Methods:To divide the cell into the experimental group (anti-LewisY antibody treated group,Rg3 treated group, combined anti-LewisY antibody and Rg3 treated group),and control group (A431) four groups. To compare the tumor control rate of each group by MTT;to drow the tumor control rate curve of each group according to MTT;to detect the expression of bcl-2 and bax in each group by immunofluorescence and Western-blot.Results:1) The cell growth speed of each experimental group was slower than the control group,and the speed of the combined anti-LewisY antibody and Rg3 treated group was the most slowest,which depended on the cell growth time and drug-dose (P<0.05);2) in 48 hours,the inhibitory effect of three experimental groups were different,the effect of combined anti-LewisY antibody and Rg3 treated group was the most obviously (P<0.05).It showed that there was synergistic effect when combined anti-Lewis Y antibody and Rg3 than use anti-Lewis Y antibody or Rg3 respectively on A431 cell;3) the expression of bcl-2 on the experimental groups were significantly down-regulated compared with the control group. Comparing with the combined group and the other three groups respectively, the expression of bcl-2 was the most down-regulated.4) In contrast,the expression of bax on the experimental groups were significantly up-regulated compared with the control group and the combined group was the most up-regulated.Conclusion:Combining Rg3 and anti-LewisY antibody can inhibit the cell lines A431 growth as well as reduce tumour cell apoptosis.The effect was more significant when use Rg3 and anti-Lewis Y antibody together than use them respectively.while they also down-regulated the expression of bcl-2 and up-regulated the expression of bax.
