Objective To investigate the correlation between the single nuclertide polymorphism in the promoter region of interleukin-10(IL-10) gene and the clinical phenotypes of hepatitis B virus(HBV)infection in Chinese population。Methods A retrospective case-control study design was applied for the present study. Using genomic DNA as templates, The genotypes of-1082 and-592 loci in the promoter region of IL-10 were analysis by polymerase chain-restriction fragment length polymorphism method (PCR-RFLP) in 175 cases of patients with hepatitis B and 153 cases of the control.And Preliminary evaluate the association between the genotypes of-1082 and-592 loci in the promoter region of interleukin-10 polymorphisms with hepatitis B virus infection consequence.Chi-square test (χ2 test) was used for statistical.Results Genotype AA,AC,CC in loci-592 in the promoter region of IL-10 were found in both patients group and the control group from Chinese Han population of Shanxi, and the genotypes distribution in two groups has no significant difference (P>0.05). The-1082AA and-1082AG genotype in IL-10 promoter region were only found in the group of patients with hepatitis B, but-1082 A A,-1082AG and-1082GG genotypes were all found in the control group. the genotypes distribution in two groups has no significant difference.But the distribution frequences of-1082A allele and-1082AA genotype in IL-10 promoter region were significantly higher in the group of patients with chronic hepatitis B than the group of patients with acute hepatitis B and the group of patients with sever hepatitis B (P 0.001,0.05)。Morever,the distribution between the group of Cirrhosis and the group of acute hepatitis B has significant difference(p=0.020).Conclusion 1.IL-10-592A/C and IL-10-1082A/G Single nucleotide polymorphism with the occurrence of hepatitis B has no significant difference.2.The-1082A allele and -1082AA genotype in IL-10 promoter region may be associated with the chronic hepatitis B infection.
