| Objective To establish a efficient method of the Wistar rat model of chronic abacterial prostatitis (CABP) and study the function of voltage-dependent calcium channels (VDCC) in the pathogenesis of chronic abacterial prostatitis.Methods Fourty male Wistar rats were divided into the nomal control group and the chronic abacterial prostatitis group stochastically, castrated and injected estradiol [0.25mg/2ml·kg-1,qd×30d] into the subcutaneous for 30 days, to establish a Wistar rat model of chronic abacterial prostatitis (CABP). And the rats of the nomal control group were injected physiological saline into the subcutaneous for 30 days. When established the model of chronic abacterial prostatitis successfully, took the protate of the two groups rats on the condition of asepsis, partial tissues was made paraffin section, hematoxylin and eosin stain, and partial tissues was abstracted the total RNA. Examined and quantitated theα1C,α1D,α1E,α1G,α1H mRNA expressions in the protate of the nomal control group and the chronic abacterial prostatitis group using reverse transcription polymerase chain reaction (RT-PCR).Results By the above treatment, 100% of the Wistar rats of the model group occurred chronic abacterial prostatitis, and none of the nomal control group. All the protate of rats expressedα1C,α1D,α1E,α1G,α1H mRNA. Compared with the nomal control group, the expressions ofα1C,α1D,α1G,α1H mRNA declined and there was statistical significance between the expressed differences ofα1G (0.30±0.27) andα1H (1.45±2.30) mRNA in the chronic abacterial prostatitis group (P<0. 05). The expressions ofα1E were hardly any, and there was no change between the two groups.Conclusion The method of castrated and injected estradiol [0.25mg/2ml·kg-1, qd×30d] into the subcutaneous can establish a Wistar rat model of chronic abacterial prostatitis successfully, and had researchful value. The protate of rats expressed L,T and R types VDCC. The abnormal expression of T-type VDCC'mRNA may be one of the causes of chronic abacterial prostatitis. |
