Objective:The goal of project was to determine whether autologous myoblast injection into the thyroarytenoid(TA) muscle after recurrent laryngeal nerve (RLN) injury could attenuate TA muscle atrophy and enhance reinnervation of motor endplates .Methods:To set up the model of UnilateralRLNtransaction were performed by microreseceing procedure in twenty rabbits. sternocleidomastoid muscle (approximately 10g) biopsies were performed in ten rabbits. Biopsies were used to create myoblast cultures for the experimental group , enzyme digestion was adapted to separate muscle satellite cells under sterile condition. Whenthe concentration reached 1×106/ml, they were transplanted. 106 autulogous myoblasts were injected into the denevated TA of each study animal,with saline injected into controls.During the cell culturing, differentiation and immunohistochemistry identification and anti-SMA were proformed. After a survival time of 7 days and 2 mouths, paraffin slides were made in transverse section orientation. HE staining and Acetylcholinesterase staining was preformed. Motic Advance 3.2 graphic analytical system was used to measure the cross section area By comparing the damaged and healthy parts of each individual,, both experimental group and control group were measured for the remnant rate of denervated muscle wet weight and the remnant rate of cross section area. The data were shown with average value and standard deviation, SPSS 11.5 statistic software was used for t test for both group..results: At 2and 8 weeks postoperatively, The group of skeletal muscle satellite cells transplanted when two weeks and eight weeks denervated thyroarytenoid muscle wet weight remnant rate(the experimental group:75.58%±7.39%及45.07%±6.88%,the control group: 55.53%±1.52%及26.89%±3.04%) and muscle cross- sectional areas of fiber remnant rate(the experimental group:72.55%±8.12% and 58.96%±6.07%;the control group: 50.23%±4.76% and 33.63%±4.41%) were better than in the control group respectively ( P< 0.05) .Conclusion:Autologous myoblast therapy may be a future treatmene for vocal cord paralysis,with current fingings demonstrating myoblast survival with attenuation of TA muscle stem cells.
