【Objective】To investigate expression of Par3, Par6 and aPKC in gastric carcinoma, and to explore the relationship between expression of Par3, Par6 and aPKC and the invasion and metastasis of gastric carcinoma.【Methods】153 gastric carcinoma samples were collected from the First Affiliated Hospital of Fujian Medical University from October 2008 to June 2009 and were made tissue microarray; by using immunohistochemistry, the expression of Par3, Par6 and aPKC was detected in different sites of gastric carcinoma (including the area of the gastric carcinoma in gastric mucosa, the center area of the gastric carcinoma and the invasive front of gastric carcinoma)and the lymph node metastasis, using normal gastric mucosa as controls.【Results】1. The expression of Par3 in different sites of gastric carcinoma was lower than in the normal gastric mucosa (p<0.01); there was no relationship between the expression of Par3 in the area of the gastric carcinoma in gastric mucosa and clinical conditions, such as age, gender, the invasion depth and lymph node metastasis, but the expression of Par3 was different in different kinds of gastric carcinoma, for example, the expression of Par3 was lower in gastric phenotype gastric carcinoma than in intestinal phenotype and mixed phenotype gastric carcinoma(P<0.05).2. In the progression gastric carcinoma, the level of expression of Par3 did not change from the area of the gastric carcinoma in gastric mucosa to the invasive front of gastric carcinoma (P>0.05); in the gastric carcinoma with lymph node metastasis, there was no different of the expression of Par3 between in primarily and in metastasis gastric carcinoma (P>0.05).3. The level of expression of Par6 in different sites of gastric carcinoma was significantly lower than in the normal gastric mucosa (p<0.01); there was no obviously relationship between the expression of Par3 in the area of the gastric carcinoma in gastric mucosa and clinical conditions(age and gender),the rate of down-regulation of Par6 in gastric phenotype carcinoma was obviously higher than that in intestinal and mixed gastric carcinoma(P<0.01),the rate of down-regulation of Par6 in gastric carcinoma which invasion to ectoptygma and out of ectoptygma was obviously higher than in gastric carcinoma which located at mucosa and under mucosa(P<0.01),and the rate of down-regulation of Par6 in gastric carcinoma with lymph node metastasis was obviously lower than that with no lymph node metastasis(P<0.01).4. During gastric carcinoma invasion and metastasis, the rate of down-regulation of Par6 did not change (P>0.05).5. Expression of aPKC in different sites of gastric carcinoma was significantly lower than in the normal gastric mucosa (p<0.01); there was no relationship between the expression of aPKC in the area of the gastric carcinoma in gastric mucosa and clinical conditions, such as age, gender, the invasion depth and lymph node metastasis, the expression of aPKC was obviously lower in gastric phenotype gastric carcinoma than in intestinal phenotype and mixed phenotype gastric carcinoma(P<0.01)6. The expression of Par3 was not associated with Par6 in the invasive front of gastric carcinoma and the lymph node metastasis, but in other site they display positive correlation with each other.【Conclusion】1 The expression of Par3 in gastric carcinoma was obviously lower than in the normal gastric mucosa, which suggested that the down-regulation expression of Par3 may be associated with carcinogenesis of gastric carcinoma; the expression of Par3 was different in different kinds of gastric carcinoma, Par3 may be associated with tissues differentiation of gastric carcinoma.2 The level of expression of Par6 in gastric carcinoma was obviously lower than in the normal gastric mucosa, it is demonstrated that the down-regulation of Par6 may be associated with carcinogenesis of gastric carcinoma; the expression of Par6 was different in different kinds of gastric carcinoma, Par6 may be associated with tissues differentiation of gastric carcinoma.3 The down-regulation of Par6 may promote the invasion and metastasis of gastric carcinoma.4 The level of expression of aPKC in gastric carcinoma was lower than in the normal gastric mucosa, which demonstrated that the down expression of aPKC may be associated with carcinogenesis of gastric carcinoma; the expression of aPKC was different in different kinds of gastric carcinoma, aPKC may be associated with tissues differentiation of gastric carcinoma.5 The down-regulation expression of Par3, Par6 and aPKC promote the carcinogenesis and progression of gastric carcinoma.
