Object:To investigate the expression of Pin1 and Cyclin D1 in primary hepatic carcinoma (PHC) and to discuss its roles in oncogenesis of PHC and relationship with clinicopathological features.Method:Forty eight hepatocancinoma specimens and corresponding adjacent tissues were collected at the surgical and eight normal liver tissues were examined by real-time quantitative reverse transcript polymerase chain reaction (RQ RT-PCR) analysis at the level of RNA , and correlation of its expression level with the clinicopathological features were analyzed by statistical method.Result: The△Ct of Pin1 expressed in PHC tissues, adjacent liver tissues and normal liver tissues is 3.01±0.74, 5.43±1.00 and 6.09±1.22.The expression level of Pin1 mRMA in the PHC tissues were significantly higher than that in the adjacent liver tissues(△Ct 3.01±0.74 vs 5.43±1.00; P<0.05) and normal liver tissues(△Ct 3.01±0.74 vs 6.09±1.22;P<0.05). The△Ct of Cyclin D1 expressed in PHC tissues, adjacent liver tissues and normal liver tissues is 4.58±0.90, 6.76±0.84 and 6.97±1.12. The expression level of Cyclin D1 mRNA in the PHC tissues were also significantly higher than that in the adjacent liver tissues(△Ct 4.58±0.90 vs 6.76±0.84; P<0.05) and normal liver tissues(△Ct 4.58±0.90 vs 6.97±1.12; P<0.05). Pin1 overexpression positively correlated with an increase in cyclin D1 levels.However, their overexpressions have no clear correlation with sex, age, clinical stage, size,protal vein thrombosis, fibrous capsule and AFP.Conclusion: Pin1 and Cyclin D1 overexpress in PHC tissues , and which might be a critical event in oncogenesis of PHC. Pin1 and Cyclin D1 may play a key role in oncogenesis of primary hepatic carcinoma (PHC) and will be a new therapy target for PHC.Their overexpressions have no clear correlation with clinical feathure.
