Hyperplasia of mammary glands is a common breast disease,the incidence is accounted for the top in varity of breast diseases.Clinical dates indicate that hyperplasia of mammary glands accounted for 65%-70% in breast diseases. Such changes generally affect premenopausal women between 20 and 50 years of age. The most common presenting symptoms are breast pain and tender nodularities in breasts.In recent years,with the accelerated pace of people's lives,work pressure increased,the incidence increase year by year,and the onset age tend to being younger.The discussion of the etiology and pathogenesis of hyperplasia of mammary glands mostly focuses on the endocrine disorders.Breast as a part of the female reproductive system, its development and function under the direct control of endocrine glands, and the main impact comes from the ovary and anterior pituitary. At the same time, breast is also indirectly regulated by the cerebral cortex. Most scholars believe that hyperplasia of mammary glands is caused by ovarian dysfunction,the increase in estrogen and reduction in progesterone,or imbalance between estrogen and progesterone.Many studies have found that the ER and PR is over-expressed in hyperplastic breast tissue,so the receptor over-expression is considered to be related to the occurrence of breast hyperplasia.Traditional Mongolian medicine Mammary-Ⅰis a traditional recipe for treatment of breast hyperplasia.After the repairment by Mongolian scholars, the prescription becomes more complete and perfect.Application of the Mammary-Ⅰhas treated nearly 2,000 patients, and received a satisfactory result.The experimental animals are adult female virgin Wistar rats. The rats were randomly divided into normal control group, model control group, low-dose treatment group, high-dose treatment group and positive drug group.In addition to normal control group,The remaining four groups were injected with estradiol (0.5mg/kg) once a day for twenty-five days,and then injection with progesterone(4mg/kg) once a day for five days induced hyperplasia of mammary glands model.The low and high dose groups were treated with Mammary-Ⅰonce a day for one month, and the positive drug group with tamoxifen.Every weekend we measured the diameter of the second pair of rat nipples during the experiment.In the end of the experiment,we killed all rats and separated serum from blood. Then determined estradiol(E2), prolactin(PRL),follicle stimulating hormone(FSH),luteinizing hormone(LH),progesterone(P) and testosterone (T) in serum.We also took breast tissue for pathological examination,and detected the expression of ER,PR protein by Immunohistochemistry and Western Blotting.End of the experiment for each of the experimental data were statistically analyzed, we got following results:1.The the nipple diameter of rats in model group is higher than normal control group (P<0.01).After treatment, the nipple diameter of low-dose,high-dose and TAM group are smaller than model group (P<0.01).The nipple diameter of all treatment groups compared with normal control group shows no significant difference (P>0.05).2.E2 in serum of model group is much higher than normal group(P<0.01).The low dose,high dose of Mammary-Ⅰand TAM can make E2 lower than model group (P<0.01),and all group treated with Mammary-Ⅰcomparing with normal group shows no significant difference(P>0.05). P and T in model group is much lower than normal group(P<0.01).After treatment by Mammary-Ⅰ,P and T increased (P<0.01),and show no significant difference comparing with normal group (P>0.05).However, treated with TAM makes P and T further decreasing. Mammary-Ⅰcan also significantly reduce LH in the mammary hyperplasia rats (P<0.01),adjust the disorder status between LH and FSH.3.Immunohistochemistry and Western Blotting results showed that the expression of ER,PR protein in model group is higher than normal group(P<0.01). Mammary-Ⅰcan reduce the ER,PR expression levels of hyperplasia breast tissue (P<0.01 or P<0.05).Through the above results, we can draw the following conclusions:1. Mammary hyperplasia model which induced by Co-injection of exogenous hormone can led to endocrine hormone disorder,mainly reflected in estrogen/progesterone imbalance and the mighty increase of ER,PR expression in breast tissue.2. Mammary-Ⅰcan effectively improve the situation of breast hyperplasia, correct endocrine disorder, make estrogen/progesterone back to normal,and reduce ER,PR expression in breast tissue.3. Mammary-Ⅰhas better effect compared with Tamoxifen, it is suitable for long-term application. Mammary-Ⅰhas higher clinical value on the treatment of breast hyperplasia.
