| Tanshin (Salvia miltiorrhiza Bge.) is a Labiatae family plant. It can activate blood circulation to dissipate blood stasis and regulate the flow of qi to alleviate pain. It is one of the most important Chinese traditional medicines. The major components of Tanshin are fat-soluble Tanshinones,as well as water-soluble Protocate-chualdehyde and Danshensu. Demand for Tanshin medicaments has increased in recent years, and one of the key bioactive components Tensioned-ⅡA has also increased rapidly. Pharmaceutical industry is very concern about the study on extraction techniques of Tanshin.Established test method of Tensioned-ⅡA content with TLC. Compare with the HPLC, TLC make the test convenient, fast and accurate. The average recovery of Tensioned-ⅡA was 99.8%, and the RSD was 2.15%. The TLC could be the test method of Tensioned-ⅡA content in the study of tanshinone extraction. It provided theoretical basis for the test of Tensioned-ⅡA content from other medicines.The traditional ethanol extraction from Tanshin was optimized by using orthogonal test method. The optimum extracting conditions were six times quantity of 70% ethanol,three times of extraction,2h a time. The rate of Tanshinone-ⅡA extraction was 82.1%, The content of Tanshinone-ⅡA was 1.49%. The content of Tanshinone-ⅡA was up to 19.23% by using 8% Na2CO3 to wash.Extracted Tanshinone from Tanshin material by SFE technology. Various factors that affect the extraction effect were investigated, Tanshinone-ⅡA as an index constituent.The optimum conditions for SFE-CO2 were obtained through orthogonal experiment, the results was as followed:soak the matrix by 95% ethanol for one hour, Pressure 25MPa, temperature 45℃, the rate flow of CO225L/h, time 3h,95% ethanol (contrast to matrix 3:1) serve as Entrainer. The rate of Tanshinone-ⅡA extraction was 84.9%. The content of Tanshinone-ⅡA and Cryptotanshinone was up to 53.56%. Compared with the technology of traditional ethanol extraction, SFE-CO2 technology was proved to be efficient. Mean while the extractum of SFE-CO2 has light color and perfect quality.Separated Tanshinone-ⅡA and cryptotanshinone from overall Tanshinones by column chromatography. This work provided a reference for the industrial process and a theoretical basis for the development and use of Tanshin. |
