Both Coptis and Phellodendron are widely used in clinic, which have the function of acesodyne,diminish inflammation and detumescence. They are made into topical preparations, such as ointment, lotion and rubber pasty, but these traditional formulation,have many insufficiencies, such as inconvenient,multiple smear, dose volume of volatile, easily contaminated clothing, and other allergic reactions. This research intended to develop a new transdermal delivery system shuanghuang cataplasm,exploring release and transdermal absorption mechanism in vitro,and promoting the development of Chinese medicine.The research mainly include the following aspects:1,Study of the extraction processing of Coptis and PhellodendronThe L9(34)orthogonal test method was used to optimize the extraction efficiency.The best extraction condition is as follows:70%(alcohol concentration),10 times(the solid-to-liquid ration),3 times(the times of extraction), lhr per time(the extraction time).2,Study of the matrix prescriptionThe L9(34)orthogonal test method was used and tacking strength,cohesive strength and appearances were used as evaluation indices.The optimal matrix prescription of shuanghuang cataplasm is NP-700: PVPK-90:CMC-Na : Polyplasdone XL-10:PVA: Kaolin:Glycerol: Dihydroxyalumimun aminoacetate: Tartaric acid=3.00:1.00:0.5: 0.5:0.25:2.5:15:0.08:0.16. The sequence of adding matrix was mixed with fills and glycerole,and then added adhesive agent, tartaric acid, and drug was added into glycerole.Matrix was slowly mixed into an uniform state,then spread on fabric and dried in room temperature5,Quality evaluation researches on preparationIn the research on the quality standard, Coptis and Phellodendron are qualitatively tested by TLC, which show the two kinds of herb medicine with good resolution and specificity.The drug contents,the weight differences and shaping were accorded with Pharmacopoeia of the People's Republic of China of 2005 edition. For adhesion experiments, the result shows that it can stick on the 24th ball (weight 19.1g), and the strength time is 425s.The permeation tests through rat skin in vitro demonstrated that the shuanghuang cataplasm exhibited zero-order kinetics equation. The drug release from the shuanghuang cataplasm according to the Higuchi dynamical equation.
