| Cerebral Palsy(CP) is an inborn brain disorder syndrome or occurred during perinatal period, characteristic of disorder of centrokinectis and abnormal posture, mainly acoompanied with multi-disabilities such as mental retardation, epilepsy, langurage disorder and audiovisual abnormality. CP is the most familiar cause of disabilities in children, which threatens their health phsically and mentailly and also places a huge burden on families and society. The treatment is still a puzzle and hotspot in neurology. We have gained great progress in epidemiology, etiology and pathology, while the treatment is mainly composed of function training which can only improve partial symptoms limitedly.Objective:To study the mechanisms on improvement of motor disability and learning and memory impariment of young rat model with CP treated by acupunture in ways of recording weight change, AbilityTest on bahavior-motor, and study-memory, and changes of oxidation-antioxidation factors in brain tissue of young rats.And to find a new method to inprove CP children's abilities on motor and study-memory through acupunture,Methods:The rats were divided randomly into normal group, Sham operation group, model group, acupunture group, moxibustion group and acu-moxibustion group.The CP model were produced by ligating left common carotid artery and hypoxia. Rats in acunpunture group were handled by rapid needling and letting a little of blood out, no retaining. QUCHI and ZUSANLI were inserted 1 fen deep, BAIHUI and temproal needle I were inserted horizontally 10 minutes. Rats in moxibustion group were treated by warming moxibustion with smokeless moxa sticks on BAIHUI and temproal needle I for 10 minutes. Rats in acu-moxibustion group were dealed with both needle and moxa sticks exactly like other groups. The normal group, Sham operation group, model group received no procedure.After trearpy, we weighed every rats and tested their behavior, then decapitated the rats and taking the coronary slices of brain tissue to record changes of the level of SOD, GSH-Px, MDA and NO.Results:Weight There was no difference among 6 groups on the first day. Mean number of gaining weights among 6 groups were different or incongruent after 18 days which was significantly(P<0.01). No statistic difference were reveled among the gaining trend of weights among the normal group, Sham operation group, moxibustion group and acu-moxibustion group, as well as between the model group, acupunture group. While the differences between the former 4groups and the latter 2 groups showed statistical significance on the gaining trend of weights(P<0.01).Muscle strength(overhanging test):Sham operation group>normal group>acu-moxibustion group>moxibustion group>model group>acupunture group The variated increases of Muscle strength produced by different treatments were statistical significance (P<0.01).Intelligence and memory (Y-maze test):normal group>Sham operation group>acu-moxibustion group>moxibustion group>acupunture group>model group.Correct times were significant different(P<0.01). Histopathology of brain tissueNormal neuron number Compared to model group, the nomal neuron number in acupunture group,moxibusiton group and acu-moxi group raised significantly after treatment (P<0.05). While no obvious differences were found among these 3 groups. The normal neurons of the Sham operation group was lower than the normal group, but not statistically related (P>0.05).Levels of SOD> GSH-Px in acupunture group, moxibusiton group and acu-moxi group were elevated then the model group after treatment, which showed significantly difference (P<0.05).The normal group (P<0.01) and Sham operation group (P>0.05) had higher level then the model group, however, neither of then showed significant difference.Compared to the model group, levels of MDA, NO in acupunture group, moxibusiton group and acu-moxi group were decreased after treatment, which showed significantly difference (P<0.05).The normal group (P<0.01) and Sham operation group (P>0.05) had lower level then the model group, however, neither of then showed significant difference.Conclusion:We found that the combination of acupunture and moxibustion received more satisfied results on raising rats'weight, abilities of motor and study-memory than the moxibustion group, which revealed the duplicate effect of the combination of acupunture and moxibustion.. CP children achieved higher effects when adopting both acupunture and moxibustion. By studing the mechanisms on improvement of motor, learning and memory of young rat model with CP treated by acupunture, we concluded that the normal neuron number in brain tissue of young rats and the levels of SOD and GSH-Px were raised while the levels of MDA and NO were decreased in the acupunture group.moxibusiton group and acu-moxi group. |
