| Objectives1. Influence of HO-1/CO system on lead and the relation between HO-1 /CO system and the lead dyed Rats'hypertension.2. Antagonism role of Sodium copper chlorophyllin(CHL) and cobalt protoporphyrin(CoPP) on the lead dyed rats'hypertension.Methods30 Wistar male rats (2 months old), whose weight were 160±12g, were divided into 4 groups (control group had 6 rats, others equal had 8 rats):①control group(distilled water),②Dye lead group (0.1% PbAc),③Dye lead (0.1% PbAc)+CoPP group (5mg/kg),④Dye lead (0.1% PbAc)+sodium copper chlorophyllin group (1.5g/L). CoPP contaminates by intraperitoneal injection every two weeks. PbAc and sodium copper chlorophyllin each confect aqueous solution through free drinking. Rats of control group drank distilled water. All rats in each group are fed normally. After 5 months, we used mouse tail arterial blood pressure gauge to measure all rats' blood pressure, and collected continued 24 hours urinary protein of urine and NAG. Then, aorta, pulmonary artery, renal artery, heart, liver and kidney were isolated. Blood lead, blood ZPP, HO-1 blood enzyme, Hb, carbon monoxide hemoglobin, organized the biopsy HO-1 immunohistochemical detection were measured. Finally, statistical analysis of all experimental data was conducted with software SPSS 1.30. Results1. Comparing with control group, Lead exposure rats's blood pressure (P<0.01) increases, so do blood lead (P<0.01) and ZPP (P<0.01); lead exposure rats's Hb (P<0.05), HbCO (P<0.01) all decrease, HO-1 blood enzyme's level decrease but (P>0.05) and HO-1 gene expression fewer.2. Comparing with control group, Dye lead+Sodium copper chlorophyllin group of blood pressure (P<0.01) increases, so do blood lead (P<0.01) and ZPP (P<0.05); Dye lead+Sodium copper chlorophyllin group's Hb decrease but (P>0.05), HbCO (P<0.01), HO-1 blood enzyme's level all increase (P<0.05) and HO-1 gene expression increase.3. Comparing with control group, Dye lead+CoPP group of blood pressure (P<0.05) increases, so do blood lead (P<0.05) and ZPP (P<0.01); Dye lead+CoPP group's Hb decrease but (P>0.05), HbCO (P<0.01)increase, HO-1 blood enzyme's level increase but(P>0.05) and HO-1 gene expression increase.4. Comparing with Dye lead group, Dye lead+Sodium copper hlorophyllin group's blood pressure (P<0.05), blood lead (P<0.05) and ZPP (P<0.05) all decrease; Hb (P<0.05), HbCO (P<0.01) and HO-1 blood enzyme's level (P<0.05) all increase and HO-1 gene expression increase.5. Comparing with Dye lead group, Dye lead+cobalt protoporphyrin (CoPP) group's blood pressure (P<0.01) and blood lead (P<0.01) all decrease but ZPP increase (P<0.05); Hb (P<0.05) and HbCO (P<0.01) all increase, HO-1 blood enzyme's level increase but(P>0.05)and HO-1 gene expression increase.Conclusion1.The HO-1/CO system may be one of reasons of causing lead dyed hypertension.2.Sodium copper chlorophyllin(CHL) and cobalt protoporphyrin(CoPP) may decrease the Lead Exposure Rats'Blood Pressure through HO-1/CO system. |
