| Objective:The purpose of this study by analyzing AGTR1 gene four SNPs,such as rs2276736,rs275650,rs5182 and rs380400 polymorphism in Chinese living in Dalian is to explore whether these SNPs are the risk factors of T2DM and hypertension.Methods:We investigated 200 T2DM patients(110 men and 90 women,mean age 55.8±12.4 yrs),211 hypertension(155 men and 56 women,mean age 55.8±10.7 yrs) and 460 normal controls(312 men and 148 women,mean age 55.8±8.3 yrs).All of them were the Han people living in Dalian.We selected four single nucleotide polymorphisms of AGTR1 gene as candidates, DNA was isolated from peripheral blood cell according to standard procedures,Sense and anti-sense primers of the four SNPs were designed according to the sequence of AGTR1 DNA.We used PCR-RFLP and HRM technique to detect these SNPs polymorphism and calculate genotype and allele gene frequency of each group,as well as differences. Univariate and multivariate logistic regression was analyzed.we analyzed the haplotypes with the haploview software.Results:1. The allele gene C frequency of rs 380400 within AGTR1 were statistically higher in the T2DM group than in the control group(P<0.05).Logistic regression analysis showed an association between the SNP and the susceptibility of T2DM in Chinese Dalian population in the rs 380400.The odds ratio of the SNP was 1.928 in an dominant model and 2.015 in a additive model after we corrected the influence of sex,age and BMI.2. The allele gene and genotype of three SNPs,such as rs2276736, rs275650,rs5182 within AGTR1 was no statistical difference between T2DM and control group(P>0.05). Univariate and multivariate logistic regression analysis showed these SNPs were not related to the genetic susceptibility of T2DM(P>0.05).3. The allele gene and genotype of four SNPs,such as rs2276736,rs275650,rs5182,rs380400 within AGTR1 was no statistical difference between hypertension and control group (P>0.05). Univariate and multivariate logistic regression analysis showed these SNPs were not related to the genetic susceptibility of hypertension (P>0.05).4..There were one haplotype block in these SNPs,and two haplotypes(TC and CC consisted of rs 5182 and rs 380400) showed significant discrepancy between case and control group (P<0.05).Conclusions:1. rs 380400 polymorphism may be associated with T2DM risk, and the C allele gene may be a genetic risk factor of T2DM in Dalian Han population.2. rs 2276736,rs 275650,rs 5182 polymorphism may not be associated with T2DM risk in Dalian Han population.3. rs 2276736,rs 275650,rs 5182 and rs 380400 polymorphism may not be associated with hypertension risk in Dalian Han population.4. Two haplotypes (the TC and CC consisted of rs 5182 and rs 380400) may be the risk factor for Type 2 diabetes in Dalian Han population. |
