| Objective:Nervous system diseases such as spinal cord injury, degenerative diseases, multiple sclerosis, motor neuron disease, mainly are due to necrosis of nerve cells. Stem cells are a class of infinite self-renewal and differentiation potential cells, they can differentiate into neuron-like cells and glial-like cells under certain conditions. They are expected to cure neurological diseases as seed cells and have raised widespread concern. Stem cells can be divided into different kinds of embryonic stem cells(ESCs) and adult stem cells(ASCs) by their different genesis. ASCs are undifferentiated cells from differentiated tissue. The advantage of ADSCs is no ethics problem when they are used as seed cells for clinical therapy, while BMSCs have been used in pre-clinical research trials because they can be obtained and purified more easily and have better neuronal differentiation. Both of ADSCs and BMSCs are easily expanded in vitro. The purpose of this experiment is to compare the differentiation potential of BMSCs and ADSCs into neuron-like cells in vitro and search the ideal seed cells for clinical stem cell transplantation.Methods:1.BMSCs were obtained by Percoll gradient centrifugation technique,and ADSCs were digested by Trypsin The phenotypically were identified by immunofluoresce staining.2. The cell viability was evaluated with cell counting and cell growth curve was generated.3.The forth-passage cells of them were induced by 10 ng/ml basic fibroblast growth factor (bFGF) for 24 h, followed by 20 ng/ml bFGF,20 ng/ml epidermal growth factor (EGF^ 20 ng/ml brain derived neurophic factor (BDNF) for 24 h,3 d,7 d,10 d,14 d. Cells were immunostained to detect the expression of neuronal markers like nestin,NSE andβ-Ⅲ-tublin.4. The number of positive cells with fluorescent staining was analyzed by the chi-square statistics.Results:1.BMSCs were obtained by pecoll density gradient centri-fugation successfully. Immunofluoresce analysis showed CD90, CD 106 were positive and CD34 were negative for ADSCs and BMSCs.2. ADSCs have a higher proliferation capacity compared to BMSCs.3. ADSCs and BMSCs can be differentiated into cells which have a fibroblastic morphology that stretched out long and thin processes.There are approximate percentages of positive cells with Nestin,β-Ⅲ-tublin, NSE, Nissl body expression between ADSCs and BMSCs.Conclusion:ADSCs have a higher proliferation capacity compared to BMSCs,however the neural differentiation capacity of ADSCs and BMSCs has no obvious difference, these results indicate that ADSCs have forpo-tential for stem cell transplantation. |
