Aim.In most Western countries, ovarian cancer is the fourth most common cause of death from cancer in women, accounting for 5% of all cancer-related deaths. Approximately one-fourth of all gynecologic malignancies are of ovarian origin, and 47% of all gynecologic cancer-related deaths are due to ovarian cancer.In our country, ovarian cancer is one of the most common pelvic genital cancer in women.The incidence of it is 2.4-5.6% in the most common tumor in women.And 22.9% of the most common pelvic genital cancer is ovarian cancer. Ovarian epithelial tumor is the the most common ovarian cancer in the date of the world. Approximately 50%-70% of all tumor of ovarian origin are ovarian epithelial tumor. The main reason for such a high mortality rate associated with this tumor is the absence of symptoms in the majority of women with early stages of the disease.70% of women present with symptoms when the disease is already at an advanced stage. In spite of significant advances in surgery and the use of new, more effective chemotherapeutic regimens, the 5-year survival of patients in all stages of the disease is only 30%-40%. The prognosis and survival of patients with ovarian tumor depends on cancer invasion, proliferation, lymph metastasis and the histopathological characteristics of the tumor.So look for the expression of many genes as to improve the early diagnosis of ovarian cancer, prognostic indicator, has become the focus of scholars who research topic.HER-2/neu (C-erbB-2) gene is a cellular oncogene. Under normal circumstances, the gene in non-activated participate in the regulation of cell differentiation. When the subject after the in vivo role of certain factors, the abnormal structure or expression is regulation. So the gene is activated, and has transforming activity with tumor. Activated HER-2/neu cancer gene takes part in cell growth and regulation,and promotes cell proliferation and growth of cells cancerous. The cell that HER-2/neu protein overexpressed has selective growth advantage.P53 gene is currently the most extensive in-depth study of the cancer gene. About 50% of human tumor are related to the gene. p53 is a tumor suppressor gene located on theshort arm of chromosome 17. P53 genes has two kinds(wild-type p53 and mutant-type p53). wtp53 may be involved in cell cycle regulation, in maintaining cell growth, inhibition of tumor proliferation play an important role. When it transformed into mtp53, then the loss of inhibition of cell growth, promote cell transformation and the excessive proliferation, resulting in tumorigenesis. Many studies have confirmed that, HER-2/neu and p53 expression is related to adenocarcinoma,including gastric cancer, ovarian cancer, breast cancer, cervical cancer and colorectal cancer and other cancer, and related to tumor invasion, stage and lymph node metastasis and other clinicopathological features.The objective of our study was to examine the expression of HER-2/neu and P53 protein in ovarian serous tumors by. immunohistochemical method, to study the relationship between HER-2/neu and P53 protein and clinical stage, pathological stage and lymph node metastasis at the protein level,and to investigate its occurrence in serous ovarian cancer and the role of relevance in determining the severity and prognosis of the role and significance.Materials and Methods88 cases of archive of resected specimens of wax block,68 cases of them are ovarian serous cancer,and 20 cases of them are ovarian serous cystadenoma as antitheses.There is no statistical difference between the age of two groups.patientswere staged according to the criteria of the International Federation ofGynecology and Obstetrics (FIGO) staging system (â… -â…£).10 cases are stageâ… ,17 cases are stage II,33 cases are stageâ…¢and 8 cases are stageâ…£.All the patients were not treated by chemotherapy, radiotherapy and sex hormone therapy before operation.Using monoclonal antibody immunohistochemical streptomycin resistance biotin protein peroxidase notation detection method (S-P) detected HER-2/neu and P53 protein expression.Paraffin sections were conventional dewaxing then to hydrate.The PeroxidaseBlocking Solution blockde the endogenous peroxidase activity,then incubated for 10 minutes at room temperature. Microwave was applied to pretreatment tissue biopsies.Then do the experiment by the SP immunohistochemistry kit instructions.The date was analyzed by SPSS 11.5 statistical software.Result1.The expression rate of HER-2/neu and P53 protein (48.53%; 55.88%) in ovarian serous cancer was significantly higher than those in ovarian serous cystadenoma.The difference were significant between the two groups(p<0.05). A positive correlation was observed between them.2.The expression rate and degree of HER-2/neu and P53 protein in advanced clinical stage was significantly higher than those in early clinical stage.The difference were significant between these groups(p<0.05).3.There was no significant association could be found between the expression of HER-2/neu and P53 protein in the age and pathological grade,and there were no significant difference (p>0.05), but there was significant association in the level of serum CA125 and there are significant difference (p<0.05). Serous ovarian carcinoma P53 and HER-2 /neu protein expression in lymph node metastasis positive group was significantly higher than without lymph node metastasis between the two groups was significant difference (P<0.05).4.Serous ovarian carcinoma P53 and HER-2/neu protein expression was common 66.18%. A positive correlation was observed btween P53 protein and HER-2/neu protein expression by the statistical analysis. Conclusion1.HER-2/neu and P53 in serous ovarian cancer positive rate was significantly higher than ovarian cystadenoma. The expression of them was associated with the occur of ovarian serous cancer.2.This showed that the expression of HER-2/neu and P53 protein was related to ovarian serous cancer stage. And the higher stage serous ovarian cancer was the higher P53 and HER-2/neu protein expression rate was, Positive correlation was observed between P53 and HER-2/neu protein expression and ovarian serous cancer stage.3.HER-2/neu and P53 protein in serous ovarian carcinoma was positively correlated with lymph node metastasis and serum CA125 level, suggesting the expression of HER-2/neu and P53 protein in serous ovarian cancer malignancy and poor prognosis.4.P53 protein expression in serous ovarian cancer with positive expression of HER-2/neu protein was higher than that in negative group, with good correlation.This suggested that joint detection reflect the combined detection of serous ovarian cancer biological behavior.
