Objective:To investigate the effects of adiponectin(ADPN)on cell proliferation and cell apoptosis and the expression of T-cadherin in human renal tubular epithelial cell cultured with high concentration of D-glucose and to illuminate the potential protection of ADPN on the kidney.Methods:①The human renal tubular epithelial cells were cultured with 5.5 mmol/L glucose (control group),30.0 mmol/L glucose (high glucose group),30.0 mmol/L glucose and concentration of 2.5μg/ml adiponectin (ADPN group), respectively. After culture for 24 h,48 h,72 h, the proliferation activity of human renal tubular epithelial cell was observed by MTT.②The human renal tubular epithelial cells were cultured with 5.5 mmol/L glucose (control group),30.0 mmol/L glucose (high glucose group),30.0 mmol/L glucose and concentration of 2.5μg/ml adiponectin (ADPN group), respectively. After culture for 24 h,48 h,72 h, the apoptotic of human renal tubular epithelial cell was observed by used the enzyme-linked immunosorbent assay (ELISA) which can measure the Caspase-3 content of the cell supernatant.③The human renal tubular epithelial cells were cultured with 5.5mmol/L glucose (control group),30.0mmol/L glucose (high glucose group),30.0mmol/L glucose+2.5μg/ml adiponectin (ADPN group), respectively. After culture for 24h,48h,72h, the expression of T-cadherin mRNA was observed by RT-PCR.Results:①The proliferation of human renal tubular epithelial cells decreased in high glucose group (P<0.05), but increased in 2.5μg/ml adiponectin group (p<0.05).②The apoptotic of human renal tubular epithelial cells increased in high glucose group (P<0.05), but delayed in 2.5μg/ml adiponectin group (P<0.05).③With the extension of time,the expression of T-cadherin decreased in high glucose group (P<0.05),but increased in adiponectin group (P<0.05).Conclusion:The high density D-glucose could inhibit the proliferation activity of human renal tubular epithelial cells and induce the cell apoptotic. Adiponectin could promote the proli-feration activity and delay the apoptotic of human renal tubular epithelial cells. With the extens-ion of time,the expression of T-cadherin decreased in high glucose group,but increased in adip-onectin group. The result prompts:the protective effect of the diabetic kidney may be through T-cadherin mediated.
