Objective To investigate the expression of CD147mRNA and protein,MMP-2, VEGF protein in human gastric carcinoma and their relationship with clinical pathological features. To explore their significances with the invasion, metastasis and prognosis. Methods The expression of CD147 and MMP-2, VEGF proteins of 161 cases of gastric carcinoma and 40 cases of adjacent normal tissues were detected by immunohistochemistry(s-p method) on tissue microarrays. The expression of CD147 mRNA of 161cases of gastric carcinoma and 40 cases of adjacent normal tissues was examined by In situ hybridization(ISH) on tissue microarrays. Results (1) In gastric carcinoma and adjacent normal tissue the positive expression rate of CD147 protein was 81.4%(130/161)and 25%(10/40)respectively, there was significant difference between the two groups(P <0.05). The expressions of CD147 protein were closely correlated with tumor size, depth of invasion, metastasis of lymph nodes and TNM stage(P <0.05), but no association was observed with the gender, age, and pathological grades. (2) the positive rate of CD147 mRNA in gastric carcinoma was 76.4%, The expressions of CD147 mRNA was significantly related with tumor size, depth of invasion and metastasis of lymph nodes(P <0.05), but it did not correlate with the gender, age, pathological grades and TNM stage(P >0.05). (3) There was a significant positive correlation between the expressions of CD147 mRNA and CD147 protein in gastric carcinoma (rs =0.676, P <0.05); (4) The positive expression rate of MMP-2 protein was77.6%(123/161) and 27.5%(11/40) respectively in gastric carcinoma and adjacent normal tissue, there was significant difference between the two groups(P <0.05). The expression of MMP-2 protein was related to the depth of invasion, metastasis of lymph nodes and TNM stage (P <0.05), but not related to the gender,age,pathological grades, and tumor size (P>0.05). (5) The positive expression rate of VEGF protein was69.6%(112/161) and 30%(12/40) respectively in gastric carcinoma and adjacent normal tissue, there was significant difference between the two groups(P <0.05). The expression of VEGF protein was related to the tumor size, depth of invasion, metastasis of lymph nodes and TNM stage(P <0.05),but not related to the gender, age, pathological grades(P>0.05).(6) There was a significant positive correlation between the expressions of CD147 protein and MMP-2 protein in gastric carcinoma (rs =0.597, P <0.05); A positive correlation was also found between the expressions of CD147 protein and VEGF protein in gastric carcinoma (rs =0.499, P <0.05). Conclusion (1)The expression of CDl47 protein and MMP-2, VEGF protein in gastric carcinoma were correlated with invasion and metastasis of gastric carcinoma, and could be potential indexes of envaluating invasion and metastasis of gastric carcinoma. (2) In gastric carcinoma, CD147 protein expression is regulated at the transcriptional level. The reduction of CDl47 protein expression is correspondingly the result of the down-regulation of transcriptional level.
