| Objective:To investigate the inhibitory effects and mechanism of triptolide on human endometrial carcinoma in nude mice, and observe influence of triptolide on the growth and physiologic function of the nude mice.Method:Human endometrial cancer cell line HEC-1B was seeded in the subcutaneous and implanted in a xenograft model by using nude mice. They were randomly divided into five groups, then they were treated with 0.9% NaCl solution, high, middle, and low dose triptolide (8,4,2 ug/day each one) and DDP 40 ug/day each one for up to 10-15 days. We observed the general station of the animals, measured the weight of animal, tumor, and tumor volume, calculated the suppression rate of tumor, and drawed the growth curve of tumors and animals. The tumor specimen, heart, liver, kidneys were observed by HE staining. The expression of Bcl-2 and VEGF were detected by immunohistochemistry.Results:1. The models of human endometrial carcinoma HEC-1B in nude mice were stable and successful.2. After 15 days of treatment, the size of the xenografts in high-dose triptolide group were 894.802±81.519 mm3,compared with1771.702±114 mm3 in the 0.9% NaCl solution control group( P <0.05, With inhibition rate of 50 %) .The growth rate of tumors was the fastest and the volume of the tumor was the largest in 0.9% NaCl solution control group, and there was significant difference with high dose triptolide group, and DDP group (P<0.05).3. The supression rates of tumor in high dose triptolide , and DDP group were 50%, 50% respectively. The supression rates of tumor in high dose triptolide group was the highest, and 0.9% NaCl solution group was the lowest.4. There was no significant difference between 0.9% NaCl solution control and triptolide groups in animal weights. The animal weights of DDP group were extenuation and there was significant difference compared with 0.9% NaCl solution control group (P<0.05). The pathological changes of tumor tissue, hearts, livers, and kidneys were no obvious difference.5. The expression of Bcl-2 in 0.9% NaCl solution group was higher than the others (P<0.05). There was no significant difference among triptolide and DDP group.6. The expression of VEGF in 0.9% NaCl solution group was higher than the others (P<0.05). There was no significant difference among triptolide; and DDP group.Conclusions:1 high dose triptolide can inhibit the growth of the human endometrial carcinoma in nude mice.2 triptolide had not influence on growth and weight of animals.3 triptolide induced apoptosis of endometrial carcinoma cells by downregulating the expression of BCL-2 and inhibit the proliferation activity of tumor cells.4 triptolide can downregulate the expression of VEGF, It inhibited neovascularization of human carcinoma in nude mice. |
